人参-山茱萸化学成分分析及其对环磷酰胺所致小鼠骨髓抑制的影响 被引量:4

摘要 目的:对人参-山茱萸改善骨髓抑制的机制进行初步研究,并进一步对其化学成分进行定量分析。方法:通过腹腔注射环磷酰胺,建立小鼠骨髓抑制模型,以外周血细胞数、骨髓有核细胞数、集落刺激因子、造血祖细胞集落数、骨髓有核细胞周期和凋亡为指标,对人参‑山茱萸改善骨髓抑制的机制进行初步研究并通过高效液相色谱法对12种人参单体皂苷和莫诺苷、马钱苷进行定量分析。结果:人参‑山茱萸可有效改善白细胞、血小板以及骨髓有核细胞数,调节细胞因子水平,增强骨髓有核细胞的分化能力,解除骨髓有核细胞的周期阻滞,减少骨髓有核细胞的凋亡,其中以中、高剂量组效果显著;人参‑山茱萸中莫诺苷含量高达15.28 mg/g,其次为马钱苷,含量为5.04 mg/g,而人参中人参皂苷以Rk_(3)、F_(2)、S‑Rg_(3)、R‑Rg_(3)含量最高,分别为0.94 mg/g、0.84 mg/g、1.33 mg/g、1.08 mg/g,人参皂苷Rg_(1)、Re、Rb_(3)、Rd均无法检测到。结论:人参-山茱萸配伍可改善环磷酰胺所诱导的小鼠骨髓抑制,其机制可能与其改善骨髓有核细胞的分化、周期凋亡有关。 Objective:To conduct a preliminary study on the mechanism of Panax ginseng C. A. Mey-Cornus officinalis Sieb. et Zucc improving myelosuppression,and to further quantitatively analyze its chemical constituents. Methods:The mouse model of bone marrow suppression was established by intraperitoneal injection of cyclophosphamide. The number of peripheral blood cells,bone marrow nucleated cells,colony-stimulating factors,hematopoietic progenitor cell colonies,bone marrow nucleated cells cycle and apoptosis were used as indicators. The mechanism of improving myelosuppression was preliminarily studied and 12 ginseng monomer saponins,morroniside and loganin were quantitatively analyzed by high performance liquid chromatography. Results:Panax ginseng C. A. Mey-Cornus officinalis Sieb. et Zucc could effectively improve the counts of leukocytes,platelets and bone marrow nucleated cells,regulated the level of cytokines,enhanced the differentiation ability of bone marrow nucleated cells,relieved the cycle arrest of bone marrow nucleated cells,and reduced the apoptosis of bone marrow nucleated cells,the middle and high dose groups had the most significant effect;The content of morroniside in Panax ginseng C. A. Mey-Cornus officinalis Sieb. et Zucc was as high as 15.28 mg/g,followed by loganin with a content of 5.04 mg/g. The contents of Rk_(3),F_(2),S-Rg_(3),and R-Rg_(3) were the highest,which were 0.94 mg/g,0.84mg/g,1.33 mg/g,and 1.08 mg/g,respectively. Ginsenosides Rg_(1),Re,Rb_(3),and Rd could not be detected. Conclusion:The combination of Panax ginseng C. A. Mey-Cornus officinalis Sieb. et Zucc can improve the myelosuppression induced by cyclophosphamide in mice,and the mechanism may be related to the improvement of the differentiation and cyclic apoptosis of bone marrow nucleated cells.
