摘要
目的研究GLIS3在胃癌中的表达与预后价值,探索GLIS3在胃癌中免疫细胞浸润情况及其参与的生物学功能。方法从TCGA数据库中下载胃癌患者GLIS3基因表达谱数据及临床病理信息,比较胃癌组织和癌旁组织中GLIS3的表达差异,分析GLIS3表达与胃癌患者临床病理特征的关系。利用Kaplan-Meier法和Cox回归分析GLIS3表达与胃癌患者临床预后相关性。运用miRWALK、TargetScan及TCGA数据库,预测调控GLIS3异常表达的miRNA。利用GSEA富集分析预测GLIS3在胃癌中可能参与的信号通路。使用TIMER2.0分析GLIS3表达与免疫细胞浸润之间的关系。结果GLIS3在胃癌组织中表达明显上调,且GLIS3的表达与肿瘤组织学分级相关(P<0.05)。生存分析显示GLIS3高表达患者比低表达患者预后更差,差异有统计学意义(P<0.05),是胃癌的独立预后因素(HR=1.288,95%CI:1.004~1.653,P<0.05)。miR-204-5p在胃癌中表达明显下调,且与GLIS3表达负相关(P<0.05)。富集分析结果显示:GLIS3基因主要富集在细胞外基质(ECM)受体相互作用、钙、黏着斑等信号通路。免疫浸润分析结果显示GLIS3表达与CD4+T细胞、巨噬细胞、中性粒细胞和树突状细胞相关(P<0.05)。结论GLIS3在胃癌中表达上调,可作为胃癌的诊断和预后的标志物,且与免疫浸润有关。
Objective To investigate the expression and prognostic value of Gli-similar3(GLIS3)in gastric cancer,and to explore the immune cell infiltration of GLIS3 in gastric cancer and its biological functions.Methods The GLIS3 gene expression profile data and clinicopathological information of gastric cancer patients were downloaded from the TCGA database to compare the expression differences of GLIS3 in the gastric cancer tissue and paracancerous tissue,and to analyze the relationship between the GLIS3 expression and clinicopathological characteristics of gastric cancer patients.The Kaplan-Meier method and Cox regression were used to analyze the correlation between the GLIS3 expression and clinical prognosis of gastric cancer patients.The miRWALK,TargetScan and TCGA databases were used to predict the miRNAs regulating aberrant GLIS3 expression.GSEA enrichment analysis was used to predict the possible signaling pathways involved in GLIS3 in gastric cancer.TIMER 2.0 was used to analyze the relationship between the GLIS3 expression and immune cell infiltration.Results The GLIS3 expression was significantly upregulated in gastric cancer tissues,and correlated with the tumor histological grade(P<0.05).The survival analysis showed that patients with high GLIS3 expression had worse prognoses than those with low expression,with a statistically significant difference(P<0.05).The GLIS3 expression was an independent prognostic factor for gastric cancer(HR=1.288,95%CI:1.004-1.653,P<0.05).The miR-204-5p expression was significantly downregulated in gastric cancer and negatively correlated with the GLIS3 expression(P<0.05).The results of enrichment analysis showed that the GLIS3 gene was mainly enriched in signaling pathways such as extracellular matrix(ECM)receptor interaction,calcium and focal adhesions.The results of immuno-infiltration analysis showed that GLIS3 expression was associated with CD4+T cells,macrophages,neutrophils,and dendritic cells(P<0.05).Conclusion The GLIS3 expression was upregulated in gastric cancer,and could be used as a diagnostic and prognostic marker in gastric cancer.It was associated with immune infiltration.
