新培养箱挥发性有机化合物对鼠胚胚胎发育的影响

The effect of volatile organic compounds in new incubator on mouse embryo development

摘要 目的 研究新建辅助生殖实验室内新培养箱挥发性有机化合物(VOCs)及甲醛对小鼠胚胎体外受精发育的影响和去除培养箱内VOCs的方法。方法 根据小鼠胚胎是否在新培养箱内分为实验组和对照组,比较装修后3个月内实验组和对照组小鼠胚胎发育的关键指标:正常受精率、卵裂率、Day 2胚胎形成率、Day 3胚胎形成率、2细胞囊胚转化率的差异及不同方法对培养箱内部VOCs去除效果。结果 第一、第二个月实验组VOCs浓度显著高于对照组,差异显著(P<0.001);甲醛浓度在第一(P<0.01)、二个月(P<0.05)也显著高于对照组;第三个月两组VOCs及甲醛浓度无明显差异;第一、第二个月,实验组正常受精率、卵裂率、2细胞囊胚转化率明显低于对照组,差异具有统计学意义;培养箱内在线过滤器VOCs的去除率[(57.87±1.89)%]明显高于纯水擦拭+干烧[(25.05±3.31)%],差异有统计学意义(P<0.001)。结论 新培养箱内存在VOCs及甲醛会对小鼠体外受精及胚胎发育产生不利影响,应采取有效措施去除VOCs及有害物质,并随时监测实验室内VOCs。 Objective To study the effects of volatile organic compounds(VOCs) and formaldehyde in a newly opened incubator in a newly-built assisted reproduction laboratory on the development of mouse embryos in vitro fertilization,and found the method of removing VOCs in the incubator. Methods The mouse embryos were divided into experimental group and control group according to whether they were in the new incubator. The key indicators of mouse embryo development in the laboratory group and control group within 3 months after decoration were compared, the differences of normal fertilization rate, cleavage rate, Day 2 embryogenesis rate, Day 3 embryogenesis rate, 2 cell blastocyst conversion rate and VOCs removal effect of different methods in the incubator. Results The VOCs concentration in the experimental group was significantly higher than that in the control group in the first and second month(P<0.001). The formaldehyde concentration in the first(P<0.01) and the second month(P<0.05) was significantly higher than that in the control group. In the third month, there were no significant difference in VOCs and formaldehyde concentrations between the two groups. In the first and second months,the normal fertilization rate, cleavage rate, and 2-cell blastocyst transformation rate of the experimental group were significantly lower than those of the control group, and the difference was statistically significant, the VOCs removal rate of the online filter in the incubator was(57.87±1.89)% significantly higher than pure water wiping+dry burning(25.05±3.31)%, the difference is statistically significant(P<0.001). Conclusion The presence of VOCs and formaldehyde in the new incubator can have adverse effects on in vitro fertilization and embryo development of mice. It is necessary to take appropriate measures to remove VOCs and harmful substances, and VOCs in the laboratory should be monitored at any time.
