摘要
以间氯苯甲酸为配体,合成了配合物{[Cu(Mcba)_(2)]_(2)·(DMF)_(2)}(Mcba=间氯苯甲酸,DMF=N,N-二甲基甲酰胺),配合物结构经元素分析、IR和X-射线单晶衍射表征,该配合物属于正交晶系,Fdd2空间群,晶胞参数为a=10.6872(5)nm,b=42.366(3)nm,c=17.3115(10)nm,Z=8,D_(c)=1.518 g·cm^(-3),F(000)=3632,最终结构残差因子R_(1)=0.0375,wR_(2)=0.0917。采用紫外、荧光和粘度法对配合物和小牛胸腺DNA(CT-DNA)及人血清蛋白(HSA)的相互作用方式进行研究,结果表明,配合物与DNA的结合常数K_(b)=3.48×10^(3)L·mol^(-1),猝灭速率常数K_(q)=1.15×10^(11)L·mol^(-1)·s^(-1)。配合物对CT-DNA的荧光猝灭是静态猝灭,结合常数K_(a)=1.98×10^(3)L·mol^(-1),结合位点n=1。配合物跟HSA结合常数K_(a)=4.20×10^(3)L·mol^(-1),猝灭速率常数值K_(q)=1.16×10^(13)L·mol^(-1)·s^(-1),结合位点数n=1。{[Cu(Mcba)_(2)]_(2)·(DMF)_(2)}对胃癌细胞A549、宫颈癌细胞Hela及肝癌细胞HepG2作用48 h后细胞的IC_(50)值分别为36.52μM、20.95μM、20.11μM。
In this paper,a copper(Ⅱ)complex{[Cu(Mcba)_(2)]_(2)·(DMF)_(2)}was synthesized with m-chlorobenzoic acid as ligand.The structure of the complex was characterized using elemental analysis,infrared(IR)and X-ray single crystal diffraction.The complex belongs to orthorhombic,Fdd2 space group,with a=10.6872(5)nm,b=42.366(3)nm,c=17.3115(10)nm,Z=8,D_(c)=1.518 g·cm^(-3),F(000)=3632 unit cell parameters.The final R_(1)=0.0375,wR_(2)=0.0917.The interactions between CT-DNA(calf thymus DNA)/HSA(human serum albumin)and the complex were evaluated by UV-Vis,fluorescence spectroscopy,and viscometry.When the complex bound to CT-DNA,the binding constant K_(b)=3.48×10^(3)L·mol^(-1),K_(q)=1.15×10^(11)L·mol^(-1)·s^(-1).The complex quenched the intrinsic fluorescence of CT-DNA via static quenching with K_(a)=1.98×10^(3)L·mol^(-1),n=1.The binding constant of the complex to HSA K_(a)=4.20×10^(3)L·mol^(-1),K_(q)=1.16×10^(13)L·mol^(-1)·s^(-1),n=1.After A549,HeLa,and HepG2 cells were subjected to{[Cu(Mcba)_(2)]_(2)·(DMF)_(2)}for 48 h,the IC_(50)were 36.52μM,20.95μM,and 20.11μM,respectively.
