显微介导鳜基因镜鲤F_(1)代肌肉中蛋白质和氨基酸组成与含量 被引量:1

摘要 本研究利用显微介导远缘杂交技术,将鳜(Siniperca chuatsi)基因组DNA直接导入镜鲤(Cyprinus carpio L)受精卵内并获得F1代,测定1龄显微介导F1代鳜基因镜鲤和普通镜鲤肌肉中蛋白质和氨基酸组成与含量。结果发现,显微介导鳜基因镜鲤F1代肌肉中粗蛋白质含量为18.69%,高于对照镜鲤的17.45%;氨基酸总量为79.77%,高于对照镜鲤的74.53%,其中4种鲜味氨基酸总含量和必需氨基酸总量分别为30.07%、28.12%和32.71%、30.48%,均高于对照镜鲤。显微介导鳜基因镜鲤F1代肌肉中必需氨基酸含量(WEAA)和总氨基酸含量(WTAA)的比值为41.01%,必需氨基酸含量与非必需氨基酸含量的比值(WEAA/WNEAA)为69.54%。结果表明,显微介导鳜基因镜鲤F1代肌肉中氨基酸的组成符合FAO/WHO标准的要求,且第一限制性氨基酸含量丰富,显著高于普通镜鲤,可在蛋白质营养上对人体起到补缺作用。本研究结果为显微介导鳜基因镜鲤的商品化提供理论依据。 In this study,the mandarin fish(Siniperca chuatsi)total DNA was injected into mirror carp(Cyprinus carpio L)by introgressive hybridization of exogenous gene with microinjection.The mirror carp F1 generation of exogenous DNA with microinjection of mandarin fish genes was tested by amplified fragment length polymorphism(AFLP)molecular marker technology.The muscle protein and amino acid composition and contents of F1 generation were determined by routine analysis methods.It was found that the gene of mandarin fish were detected in genome of transgenic mirror carp by AFLP of mandarin fish,which confirmed that the genomic fragment of mandarin fish exists in transgenic mirror carp and has no found in contrast ones,then indicates that it should be feasible to transmit genetic material between two distant related species by transgenic method at molecular level.The statistical analysis revealed that there was 18.69%of crude protein,79.77%of total amino acids,30.07%of four kinds of delicious amino acids and 32.71%of essential amino acid in transgenic mirror carp F1 generation,higher than those in non-transgenic common mirror carp ones(17.45%,74.53%,28.12%,and 30.48%,respectively),with the ratio of essential amino acid content to total amino acid content(EAA/TAA)=41.01%and the ratio of essential amino acid content to non-essential amino acid content(EAA/NEAA)=69.54%in the muscle of transgenic mirror carp F1 generation.The amino acid composition in the muscle of transgenic mirror carp F1 generation was shown to be in line with the requirements of FAO/WHO standards,and abundant content of the first restricted amino acid,which was significantly higher than that in non-transgenic common mirror carp.The findings provide a theoretical basis for the commercialization of the mirror carp with microinjection of mandarin fish genes.
