摘要
目的分析环状RNA circPVT1在食管鳞状细胞癌(ESCC)组织中的表达及其与患者临床病理特征的关系,并探讨circPVT1对ESCC细胞增殖、凋亡的影响。方法收集2017年9月至2019年12月在河北省胸科医院行手术治疗的ESCC患者的ESCC组织及相应癌旁组织,共40例。采用qRT-PCR检测ESCC组织与相应癌旁组织、ESCC细胞ECA-109与正常食管上皮细胞HEEC中circPVT1的表达水平,并分析其与患者临床病理特征的关系。将ECA-109细胞转染短发夹环状RNA敲低circPVT1的表达,采用CCK-8法检测细胞增殖情况,流式细胞术检测细胞凋亡情况,Western blot法检测增殖细胞核抗原(PCNA)及B淋巴细胞瘤-2(BCL-2)蛋白表达。结果ESCC组织中circPVT1表达水平高于癌旁组织(P<0.05),且ECA-109细胞中circPVT1表达水平高于HEEC细胞(P<0.05)。ESCC患者circPVT1高表达与肿瘤TNM分期及淋巴结转移有关(均P<0.05),与患者年龄、性别、肿瘤分化程度无关(均P>0.05)。在ECA-109细胞中敲低circPVT1的表达能够促进细胞凋亡,抑制细胞增殖,同时下调细胞中PCNA蛋白表达(P<0.05),上调BCL-2蛋白表达(P<0.05)。结论circPVT1在ESCC中高表达,敲低circPVT1的表达可抑制ESCC细胞增殖、促进凋亡。
Objective To investigate the relationship between the expression of circular RNA circPVT1 and clinicopa-thological features of esophageal squamous cell carcinoma(ESCC),and its effect on the proliferation and apoptosis of ESCC cells.Methods The ESCC and corresponding adjacent tissue samples were collected from 40 ESCC patients who underwent surgery in Hebei Chest Hospital from September 2017 to December 2019.The expression of circPVT1 in ESCC tissue and corresponding adjacent tissues was detected with qRT PCR,the relationship between circPVT1 expression and clinicopathological features of ESCC was analyzed.The expression of circPVT1 in ESCC ECA-109 cells and normal esophageal epithelial HEEC cells was also detected.The circPVT1 was knocked down in ECA-109 cells.Cell proliferation was detected by CCK-8 method,apoptosis was detected by flow cytometry,and the expression of proliferating cell nuclear antigen(PCNA)and B-cell lymphoma-2(Bcl-2)protein were detected by Western blot.Results The expression level of circPVT1 in ESCC was higher than that in adjacent tissues(P<0.05),and the expression level of circPVT1 in ECA-109 cells was higher than that in HEEC cells(P<0.05).The high expression of circPVT1 in ESCC patients was related to TNM stage and lymph node metastasis(all P<0.05),but not to age,gender and tumor differentiation(all P>0.05).Knockdown of circPVT1 in ESCC cells promoted apoptosis,inhibited cell proliferation,down-regulated the expression of PCNA protein(P<0.05)and up-regulated the expression of Bcl-2 protein(P<0.05).Conclusion Circular RNA circPVT1 is highly expressed in ESCC.Knockdown of circPVT1 can inhibit the proliferation and promote apoptosis of ESCC cells.
