摘要
目的探讨N6-甲基腺嘌呤(N6-methyladenosine,m6A)修饰的LncRNA SLC7A11-AS1调控YAP1参与口腔鳞癌(OSCC)细胞侵袭与迁移的作用机制。方法生物信息学工具预测METTL3/LncRNA SLC7A11-AS1/YAP1轴的作用关系,并借助RNA pull-down实验与RIP实验进行验证。检测细胞中m6A、METTL3、LncRNA SLC7A11-AS1、YAP1的表达水平。将细胞分为si-NC、si-SLC7A11-AS1、si-SLC7A11-AS1+pcDNA、si-SLC7A11-AS1+pcDNAYAP1组。借助划痕愈合实验与Transwell实验评估细胞迁移与侵袭能力。结果 OSCC细胞中LncRNA SLC7A11-AS1表达上调(P<0.05);敲减LncRNA SLC7A11-AS1抑制了OSCC细胞的迁移与侵袭(均P<0.05);LncRNA SLC7A11-AS1正向调控YAP1,过表达YAP1部分抵消si-LncRNA SLC7A11-AS1对细胞迁移与侵袭的作用(均P<0.05);m6A相关蛋白METTL3可促进LncRNA SLC7A11-AS1在OSCC中的表达。结论 m6A修饰的LncRNA SLC7A11-AS1在OSCC中过表达,并且通过上调YAP1促进OSCC细胞的迁移与侵袭。
Objective The purpose of this study was to investigate the mechanism of N6 methyladenine(m6 A modified LncRNA SLC7 A11-AS1 in invasion and migration of oral squamous cell carcinoma(OSCC)cells via regulating YAP1. Methods The relationship between METTL3/LncRNA SLC7 A11-AS1/YAP1 axis was predicted by bioinformatics tools and verified by RNA pull-down experiment and rip experiment. The expression level of m6 A,METTL3,LncRNA SLC7 A11-AS1 and YAP1 in OSCC cells were detected. The cells were divided into si-NC,si-SLC7 A11-AS1,si-SLC7 A11-AS1+pcDNA,si-SLC7 A11-AS1+pcDNA-YAP1 group. The ability of cell migration and invasion were evaluated by wound healing assay and Transwell assay. Results The expression of LncRNA SLC7 A11-AS1 was up-regulated in OSCC cells(P<0.05). Knockdown of LncRNA SLC7 A11-AS1 inhibited migration and invasion of OSCC cells(all P<0.05). LncRNA SLC7 A11-AS1 positively regulated YAP1,overexpression of YAP1 partially offseted the effects of si-LncRNA SLC7 A11-AS1 on cell migration and invasion(all P<0.05). m6 A related protein METTL3 could promote the expression of LncRNA SLC7 A11-AS1 in OSCC. Conclusion m6 A modified LncRNA SLC7 A11-AS1 is overexpressed in OSCC,and promotes the migration and invasion of OSCC cells by up regulating YAP1.
