基于HPLC指纹图谱研究^(60)Co-γ辐射灭菌对烧伤灵酊有效成分的影响 被引量:2

摘要 目的考察^(60)Co-γ辐照灭菌对烧伤灵酊中虎杖苷、大黄素、白藜芦醇、大黄素甲醚、盐酸小檗碱5个有效成分含量的影响及辐照前后指纹图谱变化,为烧伤灵酊的灭菌方法提供参考。方法采用Agilent ZORBAX SBC_(18)(4.6 mm×250 mm,5μm)液相色谱柱,以乙腈-0.5%磷酸溶液为流动相,梯度洗脱;流速1.0 ml/min;波长265,287 nm;柱温28℃,进样量10μl,建立烧伤灵酊HPLC指纹图谱。分别选择辐照剂量0,3,6,9 kGy对烧伤灵酊进行灭菌,比较辐照前后烧伤灵酊指纹图谱及5种有效成分含量的变化。结果当辐照剂量不超过6 kGy时,烧伤灵酊的HPLC指纹图谱无显著变化,虎杖苷、大黄素、白藜芦醇、大黄素甲醚、盐酸小檗碱含量变化无统计学意义(P>0.05)。结论^(60)Co-γ辐照灭菌法可用于烧伤灵酊的灭菌。 Objective To investigate the influence of ^(60)Co-γradiation sterilization on contents of five active ingredients of Shaoshangling Tincture including polydatin,emodin,resveratrol,physcion and berberine hydrochloride and the changes of fingerprints before and after radiation sterilization,so as to provide reference for the sterilization of Shaoshangling Tincture.Methods The analysis was carried out on an analytical column Agilent ZORBAXSB-C_(18)(250 mm×4.6 mm,5μm),using acetonitrile and 0.5%phosphoric acid solution at the flow rate of 1.0 ml/min as mobile phase of gradient elution.The detection wavelengths were 265 and 287 nm,and column temperature was 28℃,and the sample size was 10μl.The fingerprints of Shaoshangling Tincture were established.0,3,6,9 kGy of irradiation was selected for sterilization of Shaoshangling Tincture,respectively.The fingerprints and the contents of five active components before and after irradiation were compared.Results When the radiation dose was less than 6 kGy,the contents of polydatin,emodin,resveratrol,physcion and berberine hydrochloride and the fingerprints showed no significant change(P>0.05).Conclusion^(60)Co-γradiation can be used for the sterilization of Shaoshangling Tincture.
