基于网络药理学探讨补肺强体方治疗小儿反复呼吸道感染的作用机制 被引量:1

Mechanism of action of Bufei Qiangti prescription in treatment of pediatric recurrent respiratory tract infection based on network pharmacology

摘要 目的:基于网络药理学方法分析补肺强体方治疗小儿反复呼吸道感染(RRTI)的作用机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)获取补肺强体方有效成分及其蛋白靶点,根据ADME筛选中药活性主体,通过Gene Cards、OMIM、TTD等平台获取主要疾病靶点,利用String数据平台进行蛋白质互作网络分析,运用Metascape数据平台进行GO功能与KEGG通路富集分析,采用Cytoscape 3.7.2软件建立"中药-共同靶点-主要通路"网络。结果:最终获得补肺强体方治疗小儿RRTI的核心成分为甘草查尔酮A、β-谷甾醇、木犀草素、芒柄花黄素等;其核心靶点为STAT3、JUN、TNF、MAPK1等蛋白成分。通过分子对接验证,大部分靶点与化合物都有较好的结合效果。富集分析结果显示,补肺强体方可能通过乙型肝炎通路、卡波西氏肉瘤相关疱疹病毒感染通路、人T细胞白血病病毒1感染通路、TNF信号通路、Toll样受体信号通路等多个途径干预小儿RRTI。结论:本研究初步揭示了补肺强体方治疗小儿RRTI的共有成分、多个靶点、具体通路的相互作用机制,为该方的临床应用提供了理论依据及分子生物学基础。 Objective: To investigate the mechanism of action of Bufei Qiangti prescription in the treatment of pediatric recurrent respiratory tract infection(RRTI) based on network pharmacology.Methods: The Traditional Chinese Medicine Systems Pharmacology platform was used to obtain the effective constituents of Bufei Qiangti prescription and their protein targets, and the main parts of the activity of the traditional Chinese medicine were screened out based on ADME.Gene Cards, OMIM,and TTD platforms were used to obtain the main disease targets;String database was used to analyze protein-protein interaction;the Metascape data platform was used to perform GO enrichment analysis and KEGG pathway enrichment analysis;Cytoscape 3.7.2 software was used to construct a “traditional Chinese medicine-common targets-main pathways” network.Results: The core components of Bufei Qiangti prescription in the treatment of pediatric RRTI included licochalcone A,beta-sitosterol, luteolin, and formononetin, and their core targets included the proteins such as STAT3,JUN,TNF,and MAPK1.Molecular docking validation showed that most targets had a good binding effect with the components.Enrichment analysis showed that Bufei Qiangti prescription intervened against pediatric RRTI via multiple pathways including hepatitis B pathway, Kaposi’s sarcoma-related herpesvirus infection pathway, HTLV-1 infection pathway, the TNF signaling pathway, and the Toll-like receptor signaling pathway.Conclusion: This study preliminarily reveals the common components, multiple targets, and specific pathways of Bufei Qiangti prescription in the treatment of pediatric RRTI and thus provides a theoretical basis and a molecular biological basis for the clinical application of this prescription.
