摘要
为优化指甲花(Impatiens balsamina)中香豆素类成分提取工艺,并进一步探究其抑菌活性,在考察提取温度、提取时间、料液比及乙醇浓度对香豆素类成分提取率影响的基础上,通过正交试验优化指甲花香豆素类成分提取工艺,并采用抑菌圈法考察指甲花香豆素类成分对大肠埃希菌(Escherichia coli)、金黄色葡萄球菌(Staphylococcus aureus)的抑菌效果。结果表明,在单因素试验的基础上,通过正交试验优化后的香豆素类成分最佳提取工艺为提取温度60℃,提取时间90 min,料液比1∶40,乙醇浓度70%。在此条件下,香豆素类成分提取率为0.185%。指甲花香豆素类提取液有一定的抑菌活性,在强酸或强碱条件下活性较好;大肠埃希菌的抑菌圈平均直径为11.74 mm,金黄色葡萄球菌的抑菌圈平均直径为13.77 mm。
In order to optimize extraction process of coumarins from Impatiens balsamina and explore antibacterial activity of coumarins, on the basis of effects of extraction temperatures, extraction time, solid-liquid ratios and ethanol concentrations on extraction rate of coumarins, by orthogonal experiment, extraction process of coumarins from I. balsamina was optimized and antibacterial activities of coumarins on Escherichia coli and Staphylococcus aureus were investigated by bacteriostatic zone method. Results showed that on the basis of single factor experiment, the optimal extraction process of coumarins was extraction temperature 60 °C, extraction time 90 min, solid-liquid ratio1∶40 and ethanol concentration 70% with extraction rate of coumarins 0.185%. Coumarin extract from I. balsamina had certain antibacterial activity, and the activity was better in strong acid or strong alkaline conditions. Average diameter of bacteriostatic zone was 11.74 mm for E. coli and 13.77 mm for S. aureus.
