摘要
通过MTT法、细胞粘附试验、Transwell细胞迁移和侵袭试验检测不同浓度的大蒜素对肺腺癌细胞的活性、粘附、迁移与侵袭能力的变化;(RT-qPCR)逆转录-定量聚合酶链反应检测不同浓度的大蒜素对TIMP/MMP平衡的影响。本研究中发现大蒜素可呈剂量依赖性抑制肺腺癌细胞的活性、粘附、迁移和侵袭能力。大蒜素主要降低基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)的mRNA水平,同时对于组织金属蛋白酶抑制剂-1(TIMP-1)和组织金属蛋白酶抑制剂-2(TIMP-2)的mRNA水平则相反地呈剂量依赖性增加。因此我们得出结论,大蒜素可能通过抑制TIMP/MMP平衡,从而抑制肺腺癌细胞的侵袭。大蒜素可作为肺腺癌治疗的潜在抗侵袭剂。
MTT method,cell adhesion test,Transwell cell migration and invasion test were used to detect the changes in the activation,adhesion,migration and invasion of lung adenocarcinoma cells at different concentrations of allicin;(RT-qPCR)reverse transcription-quantitative polymerization enzyme chain reaction was used to detect the influence of different concentrations of allicin on the balance of TIMP/MMP.It was found that allicin could inhibit the activation,adhesion,migration and invasion of lung adenocarcinoma cells in a dose-dependent manner.Allicin mainly reduced the mRNA levels of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9),and at the same time for tissue metalloproteinase inhibitor-1(TIMP-1)and tissue metalloproteinase inhibitor-2(TIMP-2)mRNA levels increased in a dose-dependent manner on the contrary.It could be concluded that allicin may inhibit the invasion of lung adenocarcinoma cells by inhibiting the balance of TIMP/MMP.Allicin could be used as a potential anti-invasive agent in the treatment of lung adenocarcinoma.
