摘要
目的探讨lncRNA CoroMarker、lncRNA IFNG-AS1、lncRNA AF131217.1在冠状动脉粥样硬化性心脏病(coronary atherosclerotic heart disease,CAD)患者中的表达差异及临床意义。方法入选新疆医科大学附属中医医院2019年3月~2020年9月CAD患者40例为病例组,冠状动脉正常者40例为对照组,通过qRT-PCR法检测外周血单核细胞中lncRNA CoroMarker、lncRNA AF131217.1和lncRNA IFNG-AS1表达水平,ELISA法测定外周血IL-6、TNF-α水平。采用ROC曲线分析3种lncRNA对CAD患者的诊断价值及与IL-6、TNF-α水平的相关性。结果病例组lncRNA IFNG-AS1、lncRNA AF131217.1表达水平与对照组比较,差异无统计学意义(P>0.05),lncRNA CoroMarker表达水平显著升高(P<0.05),IL-6、TNF-α水平显著升高(P<0.05)。ROC曲线分析结果显示lncRNA IFNG-AS1、lncRNA AF131217.1和lncRNA CoroMarker的AUC分别为0.360(95%CI:0.180~0.540,P=0.13)、0.445(95%CI:0.259~0.631,P=0.55)、0.960(95%CI:0.908~1.000,P=0.00)。在相关性分析中,lncRNA CoroMarker与IL-6、TNF-α呈线性正相关,r值分别为0.437(P=0.005)、0.740(P=0.000)。结论lncRNA Coro-Marker在CAD患者血清单核细胞中表达水平明显高于对照组,lncRNA CoroMarker可能作为一种潜在的诊断冠心病的血清标志物,且可能与IL-6、TNF-α等炎性因子的表达呈正相关。
Objective To investigate the effect and clinical significance of lncRNA CoroMarke,lncRNA IFNG-AS1 and lncRNA AF131217.1 in patients with coronary atherosclerotic heart disease.Methods Forty patients with coronary atherosclerotic heart disease was recruited from the Xinjiang Uygur Autonomous Region Traditional Chinese Medicine Hospital afliated of the Xinjiang Medical Universi-ty from March 2019 to September 2020.Forty patients with normal coronary angiography served as a normal control group.The relative ln-cRNA CoroMarker,lncRNA AF131217.1 and lncRNA IFNG-AS1 expression level in monocytes derived from peripheral blood was de-tected by qRT-PCR.The levels of pro-inflammatory cytokines TNF-αand IL-6 were measured by ELISA.The receiver operating characteristic(ROC)curve was used to evaluate the diagnostic value of differentially expressed lncRNA for patients with coronary artery disease(CAD).Results No significant difference in expression level of lncRNA IFNG-AS1 and lncRNA AF131217.1 was observed between the two groups(P>0.05).Compared with the normal control group,the expression level of lncRNA CoroMarker and the level of TNF-αand IL-6 were increased in patients with CAD.ROC analyses showed that lncRNA IFNG-AS1(AUC=0.360;95%CI:0.180-0.540),lncRNA AF131217.1(AUC=0.445;95%CI:0.259-0.631)and lncRNA CoroMarker(AUC=0.960;95%CI:0.908-1.000)were predictive of for patients with CAD(P<0.05).IL-6(r=0.437,P=0.005)and TNF-α(r=0.740,P<0.001)correlated positively with lncRNA CoroMarker.Conclusion Compared with the normal control group,lncRNA CoroMarker ex-pression level in monocytes derived from peripheral blood was increased in patients with CAD.lncRNA CoroMarkermay serves as a poten-tial diagnostic biomarker in patients with CAD.And it may be positively correlated with the level of IL-6,TNF-αand other inflammato-ry factors.
