缺氧诱导因子-1α低表达对肺癌A549细胞增殖及侵袭能力的影响研究 被引量:3

摘要 目的研究缺氧诱导因子-1α(HIF-1α)低表达对肺癌A549细胞增殖、侵袭能力的影响,并探讨相关机制。方法取A549细胞进行缺氧培养,用于模拟肿瘤内部低氧微环境,设置为缺氧处理组,取正常培养的A549细胞设为正常组,比较两组HIF-1α表达。取对数期A549细胞,进行HIF-1α-siRNA、HIF-1α-pLentiGFP、对照质粒pLentiGFP转染,分别设为低表达组、高表达组、对照组,取不作处理的细胞设为空白组,比较四组HIF-1α表达、增殖能力、侵袭能力、趋化因子受体(CXCR4)及上皮型钙黏附蛋白(E-cadherin)表达。结果缺氧处理组HIF-1αmRNA、蛋白相对表达量高于正常组(P<0.05);对照组、低表达组、高表达组转染效率均>75%;与空白组、对照组比较,低表达组HIF-1αmRNA、蛋白相对表达量降低,24、48、72 h MTT实验A值降低,穿膜细胞数/视野减少,CXCR4蛋白相对表达量降低,E-cadherin蛋白相对表达量升高(P<0.05),高表达组HIF-1αmRNA、蛋白相对表达量升高,24、48、72 h MTT实验A值升高,穿膜细胞数/视野增加,CXCR4蛋白相对表达量升高,E-cadherin蛋白相对表达量降低,差异均有统计学意义(P<0.05)。结论HIF-1α低表达对肺癌肺癌A549细胞增殖及侵袭能力具有抑制作用,可能通过下调CXCR4,上调E-cadherin蛋白表达实现。 Objective To investigate the effect of low expression of hypoxia-inducible factor-1α(HIF-1α)on the proliferation and invasion of lung cancer A549 cells,and to explore related mechanisms.Methods A549 cells were taken into hypoxia culture to simulate the hypoxic microenvironment within tumor.The cultures were set as a hypoxia treatment group,while the normal cultured A549 cells were set as a normal group.The expression of HIF-1αwas compared between the two groups.A549 cells in logarithmic phase were transfected with HIF-1α-siRNA,HIF-1α-pLentiGFP,and control plasmid pLentiGFP.These were set as a low expression group,a high expression group,and a control group,respectively,and cells without treatment were set as a blank group.The expression of HIF-1α,proliferation ability,invasion ability,expression of chemokine receptor(CXCR4)and epithelial cadherin(E-cadherin)were compared between each group.Results The relative expression of HIF-1αmRNA and protein in the hypoxia treatment group were higher than those in the normal group(P<0.05).Fluorescence microscopic results showed that the transfection efficiency of the control group,the low expression group and the high expression group were all>75%.Compared with the blank group and the control group,the relative expression of HIF-1αmRNA and protein,the A value of 24,48,and 72 h MTT experiment,the number of penetrating cells/field of view,the relative expression of CXCR4 protein in the low-expression group were decreased,while the relative expression of E-cadherin protein was increased(P<0.05).Compared with the blank group and the control group,the relative expression of HIF-1αmRNA and protein,the A value of 24,48,and 72 h MTT experiment,the number of penetrating cells/field of view,the relative expression of CXCR4 protein in the high-expression group were increased,while the relative expression of E-cadherin protein was decreased(P<0.05).Conclusion The low expression of HIF-1αhas an inhibitory effect on the proliferation and invasion of lung cancer A549 cells.It may be achieved by down-regulating CXCR4 and up-regulating the expression of E-cadherin protein.
