摘要
目的探讨RABIF在肝癌细胞中的表达及其功能。方法采用Real-time PCR检测30例肝癌组织以及相应正常肝组织中RABIF的表达水平,并检测RABIF在肝癌细胞系及正常肝细胞系中的表达情况。通过shRABIF转染肝癌细胞系HepG2敲低RABIF;采用CCK-8实验检测细胞增殖改变情况;采用Transwell实验检测细胞侵袭和迁移情况。结果 RABIF在肝癌组织中的表达高于正常肝组织,差异有统计学意义(P<0.05)。敲低RABIF能抑制肝癌细胞增殖,并且抑制肝癌细胞的侵袭和转移(P<0.05)。结论肝癌组织中RABIF的表达高于正常肝组织,RABIF促进肝癌细胞增殖,并且能够提高肝癌细胞侵袭转移水平,有望成为肝癌治疗的潜在靶点。
Objective To investigate the expression and function of RABIF in HCC. Methods Real-time quantitative PCR was used to detect the expression level of RABIF in 30 liver cancer tissues and corresponding normal liver tissues, and to detect the expression level of RABIF in HCC cell lines and normal liver cell lines. RABIF was knocked down by sh-RABIF transfection of Huh7. CCK-8 kit was used to detect the changes of cell proliferation. Transwell assay was used to detect the invasion and metastasis of HCC cells. Results The expression of RABIF in liver cancer tissues was higher than that in normal liver tissues(P<0.05). RABIF knockdown inhibited the proliferation of HCC cells, and the invasion and metastasis of HCC cells(P <0.05). Conclusion The expression of RABIF in HCC tissues is higher than that in normal liver tissues. RABIF promotes the proliferation of HCC cells and can improve the invasion and metastasis of HCC cells, which is expected to be a potential target for the treatment of HCC.
