不同方法检测乳腺癌循环肿瘤细胞的诊断价值研究 被引量:1

Study on the effect and diagnostic value of detecting circulating tumor cells in breast cancer patients via different methods

摘要 以一年内临床收治的乳腺癌和乳腺纤维瘤患者为研究对象,同期健康者为对照,利用微流控免疫磁珠筛选+免疫荧光检测法(Microfluidic-beads)、差异富集-荧光免疫染色原位杂交技术(SE-iFISH)、RNA原位杂交法(RNA-FISH)3种方法对研究对象外周血循环肿瘤细胞(CTCs)进行检测,比较观察组和对照组CTCs阳性检出率的差异,探究不同检测方法对乳腺癌患者的CTCs检测效果及诊断价值。结果表明:采用SE-iFISH法和RNA-FISH法检测时,观察组患者阳性检出率较高,分别为88%和100%。但采用Microfluidic-beads法时,观察组患者阳性检出率较低,仅为48%。基于CTCs的检测表征乳腺癌方面,SE-iFISH法和RNA-FISH法具有较高的灵敏度及特异性。这一研究对后期乳腺癌患者的临床诊断与治疗具有一定的指导意义。 The detection effect and diagnostic value of different detection methods on circulating tumor cells(CTCs)in breast cancer patients were investigated and compared in this work.Patients with breast cancer or breast fibroma admitted to the hospital were selected as the observation and research objects,and healthy people during the same period(one year)were selected as the control group.Three different detection methods were used to perform CTCs detection on the peripheral blood of the three groups of subjects collected,including Microfluidic immunomagnetic bead screening+immunofluorescence detection(Microfluidic-beads),Subtraction enrichment-Immunofluorescence in situ hybridization(SE-iFISH),and RNA fluorescence in situ hybridization(RNA-FISH).The difference in the positive detection rate of CTCs between the observation group and the control group were compared.Results showed that the positive rate of the observation group was high when the SE-iFISH method or the RNA-FISH method were used,which were 88% and 100%,respectively.However,the positive rate of patients in the observation group was low and only 48% when the Microfluidic-beads method was used.SE-iFISH method and RNA-FISH method had high sensitivity and specificity for detecting and characterizing breast cancer via detecting CTCs.The results in this study have certain guiding significance on clinical diagnosis and treatment of patients with breast cancer.
