摘要
目的 研究非酒精性脂肪性肝炎转变成肝癌(NASH-HCC)中的关键差异表达基因。方法 选择4对NASH-HCC和癌旁石蜡组织进行Affymetric芯片表达谱检测,生物信息学技术筛选差异表达基因,利用DAVID数据库对差异表达基因进行GO功能和KEGG信号通路分析,应用STRING数据库对差异表达基因进行蛋白-蛋白互作网络分析,应用Cytoscape软件筛选核心基因。结果 NASH-HCC癌与癌旁组织中共筛选到279个差异表达基因,GO功能富集分析发现这些差异表达基因显著富集到脂肪代谢、糖脂生物合成、脂肪酸水解及细胞凋亡等相关的功能,KEGG通路富集分析表明差异表达的基因显著与脂肪酸合成与代谢、病毒感染及有机物代谢等通路相关。蛋白相互作用因子调控网络分析发现多能干性基因(c-Myc、Oct-4、KLF-4和Nanog)和形态分化基因(Gli-1)在人NASH-HCC标本中高表达,且在PPI中处中心位置,与其他差异表达基因存在广泛的蛋白互作关系。结论 多能干性基因和形态分化基因可能在NASH-HCC病变过程中发挥至关重要的作用。
Objective To investigate the key differentially expressed genes in the transformation of nonalcoholic steatohepatitis into hepatocellular carcinoma(NASH-HCC).Methods Four NASH-HCC and adjacent paraffin tissues were selected for expression profiling by Affymetric microarray,bioinformatics technology was used to screen differentially expressed genes.Differentially expressed genes were analyzed by GO function and KEGG signaling pathway using DAVID database.Differentially expressed genes were analyzed by protein-protein interaction network using STRING database.Core genes were screened using Cytoscape software.Results A total of 279 differentially expressed genes were identified in cancerous and adjacent tissues.GO analysis showed that the differentially expressed genes were significantly enriched in the functions of fat metabolism,glucose and lipid biosynthesis,fatty acid hydrolysis and apoptosis.KEGG pathway analysis indicated that the differentially expressed genes were significantly associated with fatty acid synthesis and metabolism,viral infection and organic metabolism.Protein interaction factor regulatory network analysis revealed that pluripotent sternness genes(c-Myc,Oct-4,KLF-4,and Nanog)and morphological differentiation gene(Gli-1)were highly expressed in human NASH-HCC specimens and centrally located in PPIs,with extensive protein interaction relationships with other differentially expressed genes.Conclusion Pluripotent sternness genes and morphological differentiation genes may play a crucial role in the pathological process of NASH-HCC.
