摘要
目的:建立益心通脉颗粒HPLC指纹图谱,并测定哈巴苷、毛蕊异黄酮葡萄糖苷、阿魏酸、甘草苷、丹酚酸B、人参皂苷Rg_(1)、人参皂苷Rb_(1)、丹参酮Ⅱ_(A)的含量。方法:用75%甲醇水溶液提取该样品细粉,采用Phenomenex Luna-C_(18)色谱柱(250 mm×4.6 mm, 5μm),流动相为乙腈(A)-0.05%磷酸溶液(B),梯度洗脱;检测波长为205,260 nm,柱温:30℃,流速:1.0 ml·min^(-1),进样量:10μl。结果:16批样品中有31个共有峰,相似度均大于0.98。8个成分在试验范围内线性关系良好(r≥0.999 4),平均加样回收率为98.19%~99.33%,RSD为0.8%~1.4%(n=6)。结论:所建立方法稳定简单,指纹图谱结合多成分含量测定为益心通脉颗粒的质量评价提供了试验依据。
Objective: To establish the HPLC fingerprint of Yixin Tongmai granules and to determine the contents of harpagide, calycosin 7-O-β-D-Glucopyranoside, ferulic acid, liquiritin, ginsenoside Rg_(1), salvianolic acid B, ginsenoside Rb_(1)and tanshinone Ⅱ_(A). Methods: The sample powder was extracted with 75% methanol aqueous solution. The HPLC detection was performed on a Phenomenex Luna-C_(18)chromatographic column(250 mm×4.6 mm, 5 μm) with acetonitrile(A)-0.05% phosphoric acid(B) as the mobile phase with gradient elution. The detection wavelengths were set at 205 nm and 260 nm, the column temperature was 30 ℃, the flow rate was 1.0 ml·min^(-1), and the injection volume was 10 μl. Results: There were thirty-one common peaks in the HPLC fingerprints of sixteen batches of samples with the similarities more than 0.98. The linear relationships of 8 components were good within the test ranges(r≥0.999 4), the average recoveries were 98.19%-99.33%, and the RSDs ranged from 0.8% to 1.4%(n=6). Conclusion: The method is simple and accurate, which provides reference for the quality control of Yixin Tongmai granules.
