柴黄清胰活血颗粒提取工艺研究

摘要 目的优化柴黄清胰活血颗粒的提取工艺。方法分别采用传统水煎、粉碎-渗漉-水煎、渗漉-水煎3种提取工艺提取柴黄清胰活血颗粒中14味中药材的有效成分;采用高效液相色谱法同时测定不同提取工艺制得的柴黄清胰活血颗粒中大黄的指标性成分芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚的含量,优选提取工艺,并比较3种提取工艺的各有效成分的含量和转移率。结果采用渗漉-水煎提取工艺制得的颗粒中芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚的总含量为0.8357 mg/g,上述成分的转移率分别为74.59%,74.61%,73.88%,52.86%,53.95%,均高于传统水煎提取工艺。结论渗漉-水煎提取工艺客观可行,稳定合理,可为工业化生产提供理论依据。 Objective To optimize the extraction process of Chaihuang Qingyi Huoxue Granules.Methods The active ingredients of 14 Chinese herbal medicines in Chaihuang Qingyi Huoxue Granules were extracted by three extraction processes:traditional water decoction,pulverization-percolation-water decoction,and percolation-water decoction.The contents of aloe-emodin,rhein,emodin,chrysophanol and physcion in Chaihuang Qingyi Huoxue Granules prepared by different extraction processes were determined by the high-perfor-mance liquid chromatography(HPLC)method.The extraction processes were optimized,and the contents and transfer rates of the active ingredients extracted by three extraction processes were compared.Results The total contents of aloe-emodin,rhein,emodin,chryso-phanol and physcion in the granules prepared by the percolation-water decoction extraction process were 0.8357 mg/g,and the transfer rates of the above ingredients were 74.59%,74.61%,73.88%,52.86%and 53.95%,respectively,which were higher than those in the granules prepared by the traditional water decoction.Conclusion The extraction process of percolation-water decoction is objective,feasible,stable and reasonable,which can provide a theoretical basis for industrial production of Chaihuang Qingyi Huoxue Granules.
