摘要
成纤维细胞活化蛋白(FAP)是一种Ⅱ型跨膜丝氨酸蛋白酶,结构上不仅具有与二肽基肽酶4相似的二肽基肽酶活性,还具有独特水解和灭活成纤维细胞生长因子21(FGF-21)等其他内源性多肽酶活性的能力,这在多肽类药物的代谢中起着重要的作用。FAP在糖脂代谢紊乱人群中水平升高,并且动物研究进一步发现抑制FAP的活性可以增加循环中FGF-21的水平从而有效改善葡萄糖耐量和脂质代谢。本文对FAP在糖脂代谢中作用的研究进展进行综述。
Fibroblast activation protein(FAP)is a kind of typeⅡtransmembrane serine protease.It not only has a dipeptidyl peptidase activity similar to dipeptidyl peptidase-4,but also has the ability to hydrolyze and inactivate other endogenous polypeptidas such as fibroblast growth factor-21(FGF-21),which plays an important role in the metabolism of peptide drugs.The level of FAP elevated in people with glycolipid metabolism disorders,and animal studies have further found that inhibiting the activity of FAP can increase the level of FGF-21 in the circulation,thereby effectively improving glucose tolerance and lipid metabolism.This paper reviews the research progress on the role of FAP in glycolipid metabolism.
