摘要
运用超声辅助酶法提取红薯叶多糖,利用响应面设计对提取工艺进行优化,结果表明:红薯叶多糖最佳提取条件为纤维素酶添加量1.5%(以红薯叶粉质量为基准)、超声时间32 min、超声功率210 W,在此条件下粗多糖SPLU得率为(8.16±0.13)%。经DEAE-52纤维素柱层析、Sephadex-G100葡聚糖凝胶柱纯化,得到红薯叶纯化多糖SPL-Ⅰ、SPL-Ⅱ2个组分。通过紫外光谱、红外光谱分析得出,多糖SPL-Ⅰ、SPL-Ⅱ无核酸、蛋白质等杂质,有明显的多糖特征吸收峰,结构为吡喃环。在扫描电镜(SEM)下观察,随着放大倍数的增加,多糖表面结构致密,层次不一,可见圆球状颗粒。运用凝胶色谱法(GPC)分析得出,多糖SPL-Ⅰ、SPL-Ⅱ的重均相对分子质量为4.48×10^(4)、5.19×10^(4)。体外抗氧化试验表明,红薯叶多糖抗氧化能力由大到小为SPL-Ⅰ>SPL-Ⅱ>SPLU。
The ultrasonic-assisted enzymatic method was used to extract polysaccharides from sweet potato leaves,and the extraction process was optimized through response surface design.The results showed that the optimal extraction conditions for sweet potato leaf polysaccharides were as follows:cellulase dosage 1.5%(based on the mass of sweet potato leaves powder)and ultrasonic time 32 min,ultrasonic power 210 W.Under these conditions,the polysaccharide SPLU yield was(8.16±0.13)%.Purified by DEAE-52 cellulose column chromatography and Sephadex-G100 dextran gel column,two components of sweet potato leaf purified polysaccharides SPL-Ⅰand SPL-Ⅱwere obtained.Through the analysis of ultraviolet and infrared spectroscopy,it was concluded that the polysaccharides SPL-Ⅰand SPL-Ⅱhad no impurities such as nucleic acid and protein,and had obvious characteristic absorption peaks of polysaccharides,and the structure was a pyran ring.Observed under a scanning electron microscope(SEM),as the magnification increased,the surface structure of the polysaccharide was dense,with different levels,and spherical particles could be seen.Using gel chromatography(GPC)analysis,the weight-average relative molecular masses of polysaccharides SPL-Ⅰand SPL-Ⅱwere 4.48×10^(4)and 5.19×10^(4).The in vitro antioxidant test showed that the antioxidant capacity of sweet potato leaf polysaccharides was SPL-Ⅰ>SPL-Ⅱ>SPLU from large to small.
