脊髓5-HT_(2A)受体介导延胡索乙素对福尔马林致痛大鼠的镇痛作用 被引量:2

Spinal 5-HT_(2A) receptor mediates the analgesic effect of tetrahydropalmatine on formalin-induced pain in rats

摘要 [目的]探讨延胡索乙素(l-THP)对福尔马林致痛大鼠的镇痛作用及其机制.[方法]取清洁级雄性SD大鼠,随机分为生理盐水对照组、福尔马林模型组、布洛芬阳性对照组及l-THP高、中、低剂量组.观察l-THP对福尔马林致痛大鼠舔爪时间、L4-L5脊髓后角c-Fos蛋白及5-HT_(2A)受体表达的影响.[结果]大鼠足底注射给予福尔马林后出现双相痛的表现.与福尔马林模型组比较,布洛芬阳性对照组及l-THP高、中、低剂量组福尔马林诱导的第Ⅱ相痛时间均明显缩短(P<0.05).大鼠足底注射给予福尔马林30、60、90 min时L4-L5腰段脊髓灰质后角均有c-Fos蛋白和5-HT_(2A)受体表达,且均在60 min时表达最明显.大鼠足底注射给予福尔马林60 min后,福尔马林模型组大鼠L4-L5腰段脊髓灰质后角内c-Fos表达水平明显高于生理盐水对照组(P<0.05),布洛芬阳性对照组及l-THP高、中、低剂量组c-Fos表达水平均明显低于福尔马林模型组(P<0.05);福尔马林模型组大鼠L4-L5腰段脊髓灰质后角内5-HT_(2A)受体表达水平较生理盐水对照组明显升高(P<0.05),布洛芬阳性对照组及l-THP高、中、低剂量组5-HT_(2A)受体表达水平均明显低于福尔马林模型组(P<0.05).[结论]L4-L5脊髓后角c-Fos蛋白和5-HT_(2A)受体参与了由福尔马林诱导的机体疼痛感觉过程;l-THP对福尔马林引起的疼痛效应具有抑制作用,且中、高剂量l-THP的抑制作用较布洛芬的效果更为显著,其作用机制可能是由5-HT_(2A)受体所介导的. OBJECTIVE To investigate the analgesic effect and its mechanism of tetrahydropalmatine(l-THP)on formalin-induced pain in rats.METHODS Male SD rats of clean grade were randomly divided into saline control group,formalin model group,ibuprofen positive control group and l-THP high,medium and low dose group.The effects of l-THP on paw licking time,c-Fos protein and 5-HT_(2A) receptor expression in L4-L5 spinal dorsal horn of formalin-induced pain rats were observed.RESULTS The rats presented with biphasic pain after plantar injection of formalin.Compared with formalin model group,formalin-induced phaseⅡpain was significantly shortened in ibuprofen positive control group and l-THP high,medium and low dose group(P<0.05).C-Fos protein and 5-HT_(2A) receptor in L4-L5 lumbar spinal dorsal horn were expressed after 30,60 and 90 min of plantar injection of formalin,and the expressions were most obvious at 60 min.After 60 min of plantar injection of formalin,the expression of c-Fos protein in L4-L5 lumbar spinal dorsal horn of rats in formalin model group was significantly higher than that in the normal saline control group(P<0.05),while the expression of c-Fos protein in ibuprofen positive control group and l-THP high,medium and low dose group was significantly lower than that in the formalin model group(P<0.05).The expression of 5-HT_(2A) receptor in L4-L5 lumbar spinal dorsal horn of rats in formalin model group was significantly higher than that in the normal saline control group(P<0.05),and the expression of 5-HT_(2A) receptor in ibuprofen group and l-THP high,medium and low dose group was significantly lower than that in formalin model group(P<0.05).CONCLUSION C-Fos protein and 5-HT_(2A) receptor in L4-L5 lumbar spinal dorsal horn may be involved in the process of body pain sensation induced by formalin.L-THP has a certainly inhibitory effect on formalin induced-pain,and the inhibitory effect of l-THP at medium and high dose was more significant than that of ibuprofen,and its mechanism might be mediated by 5-HT_(2A) receptor.
