摘要
目的探讨Polo样激酶1(PLK1)以及鼠双微体基因2(MDM2)在胃癌组织的表达及其与临床病理因素之间的关系。方法选取长治医学院附属和济医院和长治市人民医院2017年4月至2021年3月病理确诊的99例胃癌组织和癌旁组织,应用免疫组织化学方法检测组织中PLK1蛋白以及MDM2蛋白表达水平,采用χ^(2)检验并结合临床资料分析PLK1蛋白和MDM2蛋白与胃癌临床病理特征之间的关系。结果胃癌组织PLK1蛋白表达率为79.80%(79/99)、对应癌旁组织PLK1蛋白表达率为12.12%(12/99),两者差异有统计学意义(χ^(2)=91.283,P<0.01),胃癌组织MDM2蛋白表达率为66.67%(66/99)、对应癌旁组织MDM2蛋白表达率为26.26%(26/99),两者差异有统计学意义(χ^(2)=32.486,P<0.01)。PLK1表达水平与胃癌TNM分期、淋巴结转移明显相关(χ^(2)=5.602、6.550,P<0.05);MDM2表达水平与胃癌TNM分期、组织学分化程度、淋巴结转移明显相关(χ^(2)=5.263、9.949、4.583,P<0.05)。结论 PLK1蛋白和MDM2蛋白过度表达可能促进胃癌恶性增殖及转移,联合检测上述两项指标有助于判断胃癌恶性程度、转移潜能和预后。
Objective To investigate the expression of polo-like kinase 1(PLK1)and murine double minute 2(MDM2)in gastric carcinoma and their relationship with clinicopathological factors and clinical significance.Methods Immunohistochemistry was used to detect the expression levels of PLK1 protein and MDM2 protein in 99 cases of gastric carcinoma tissues and paracancerous tissues diagnosed by pathology in Heji Hospital Affiliated to Changzhi Medical College and Changzhi People′s Hospital from April 2017 to March 2021,the relationship between PLK1 protein and MDM2 protein and the clinicopathological characteristics of gastric carcinoma were analyzed byχ^(2) test combined with clinical data.Results The expression rate of PLK1 protein in gastric carcinoma tissues was 79.80%(79/99)and that in adjacent tissues was 12.12%(12/99),and the difference was statistically significant(χ^(2)=91.283,P<0.01),the expression rate of MDM2 protein in gastric carcinoma tissues was 66.67%(66/99),and that in adjacent tissues was 26.26%(26/99),the difference was statistically significant(χ^(2)=32.486,P<0.01).the expressions of PLK1 were related gastric carcinoma to tumor node metastasis stage(TNM stage)and lymph node metastasis(χ^(2)=5.602,6.550,P<0.05),the difference was statistically significant.the expressions of MDM2 were related gastric carcinoma to TNM stage,histology grade and lymph node metastasis(χ^(2)=5.263,9.949,4.583,P<0.05),the difference was statistically significant.Conclusion The overexpression of PLK1 protein and MDM2 protein may promote the malignant proliferation and metastasis of gastric carcinoma,and the combined detection of these two indexes can be used as an important reference to judge the malignant degree,metastatic potential and prognosis of gastric carcinoma.
