一株耐碳青霉烯鲍曼不动杆菌的分离鉴定及6-苄氨基嘌呤对其体外抑菌作用研究

摘要 目的:分离鉴定一株耐碳青霉烯鲍曼不动杆菌(carbapenem-resistant Acinetobacter baumannii,CRAB)并研究6-苄氨基嘌呤(6-Benzylaminopurine,6-BA)在体外对其抑菌效果。方法:通过细菌形态学、16S rDNA测序和药敏试验对临床分离株CRAB-1进行鉴定,通过倍比稀释法研究6-BA对CRAB的最低抑菌浓度,通过分析CRAB的细胞膜通透性和生长曲线,探究6-BA对CRAB的抑菌机制。结果:菌株CRAB-1为耐碳青霉烯鲍曼不动杆菌,体外抑菌实验表明,6-BA对CRAB的最低抑菌浓度为256μg/mL,且随着浓度增大,抑菌作用增强。6-BA对CRAB的对数生长期和稳定期均有显著抑制作用,但对细胞膜通透性无显著影响。结论:6-BA对CRAB具有抑菌效果,为抗CRAB感染提供了新的思路。 Objective To isolate and identify a carbapenem-resistant Acinetobacter baumannii(CRAB)strain and investi⁃gate the inhibitory effects of 6-benzylaminopurin(6-BA)on CRAB in vitro.Methods The clinical isolate CRAB-1 was i⁃dentified through bacterial morphology,16S rDNA sequencing and drug sensitivity test.The minimum inhibitory concentra⁃tion of 6-BA on CRAB was studied by the doubling dilution method,and the antibacterial mechanism of 6-BA on CRAB was explored through analyzing the cell membrane permeability and growth curve.Results Strain CRAB-1 is carbapenem-resistant Acinetobacter baumannii.The MIC of 6-BA on CRAB was 256μg/mL.The inhibitory effect of 6-BA increased a⁃long with the increase of the concentration.6-BA exhibited significant inhibitory effect on the logarithmic phase and station⁃ary phase of CRAB,but showed no significant influence on cell membrane permeability.Conclusion Our preliminary study certified the bacteriostatic effect of 6-benzylaminopurin(6-BA)on carbapenem-resistant Acinetobacter baumannii(CRAB),which provides new idea for anti-CRAB infection.
