摘要
【目的】明确侵染贵州番茄的番茄黄化曲叶病毒(Tomato yellow leaf curl virus,TYLCV)分离物与其他地区TYLCV的系统进化关系,为贵州番茄黄化曲叶病毒病的防控提供科学依据。【方法】将采自贵州省关岭县的番茄疑似TYLCV样品提取DNA,用菜豆金色黄花叶病毒属(Begomovirus)通用引物BegoAFor1/BegoARev1进行PCR检测,并用TY3/TY5引物扩增并克隆TYLCV贵州分离物的DNA-A全长序列,分析序列同源性及系统进化情况。【结果】从病样中分离得到的致使贵州番茄叶片表现黄化、皱缩等症状的病原为TYLCV,通过构建系统进化树可知,TYLCV贵州分离物(TYLCV-GZ)的DNA-A与TYLCV-Mexico和TYLCV-XJKS的相似性最高,核苷酸一致性达99%以上,其与国内其他地区TYLCV分离物归属一致,属于TYLCV-IL株系,且与TYLCV-SDLC(山东)和TYLCV-XJKS(新疆)聚在一个分支上,说明其亲缘关系最近。【结论】引起贵州关岭县番茄叶片黄化、皱缩的病原为番茄黄化曲叶病毒(Tomato yellow leaf curl virus,TYLCV),且属于TYLCV-IL株系。
【Objective】The phyletic evolution relationship between Guizhou Tomato yellow leaf curl virus(TYLCV-GZ)isolate and reported TYLCV isolates in GenBank is clarified to provide the scientific basis for prevention and control of TYLCV in Guizhou.【Method】The PCR of the DNA extracted from suspected TYLCV samples in Guanling County,Guizhou Province is detected by using Begomovirus universal primer BegoAFor1/BegoARev1.The DNA-A full length sequence of TYLCV-GZ isolate are amplified and cloned by using TY3/TY5 primer to analyze the sequence homology and phyletic evolution.【Result】TYLCV-GZ is the pathogen isolated from the diseased samples,which causes the yellowing and shrinkage of tomato leaves in Guizhou.The DNA-A of TYLCV-GZ isolate has the highest similarity with TYLCV-Mexico and TYLCV XIKS according to construction of phylogenetic tree and the nucleotide consistency reaches more than 99%.TYLCV-GZ belongs to TYLCV-IL strain.TYLCV-GZ,TYLCV-SDLC(Shandong,China)and TYLCV-XJKS(Xinjiang,China)are clustered into a branch with the closest genetic relationship.【Conclusion】The pathogen causing yellowing and shrinkage of tomato leaves is TYLCV-GZ,which belongs to TYLCV-IL strain.
