摘要
CRISPR/Cas9系统通过对基因的定向编辑,对基因组编辑产生了革命性的影响,成功促进了突变体筛选在基因组水平上的新发展。虽然该系统构建突变体文库速度快,针对性强,且具有巨大的应用前景,目前在一些重要作物的研究中已取得了重要进展,但其在木本植物中的应用仍处于初级阶段。本研究总结了CRISPR/Cas9系统在植物基因编辑文库构建方面的应用,探究该系统如何应用于构建突变敲除体库。将构建基因编辑突变体文库作为深入研究基因功能的手段,本研究综述的信息将对推进木本植物相关研究提供有益的借鉴。
CRISPR/Cas9 system delivers a critical impact on genome editing by directional editing of genes,and it has successfully promoted the new development of mutant screening at the genome level.Although the mutant library constructed by this system is fast and targeted,with great application prospects,its application in woody plants is far behind that in important crops.This study reviews the application of the CRISPR/Cas system in plant gene editing library screening and explores how this system can be applied in constructing mutant knockout libraries.By using the construction of gene editing mutant libraries as a reference for in-depth analysis of gene functions,this study can facilitate relevant researches on woody plants.
