纳米药物药动学研究方法进展

摘要 基于纳米技术的纳米药物开发是全球学术界和工业界的一个热点研究领域.然而,纳米药物安全风险(毒性)方面的监管政策尚未得到很好的确立.主要挑战是缺乏有效的分析方法来准确评估纳米药物在复杂生物系统中各形态(游离态,结合或包裹态)的浓度.目前纳米药物药动学的研究思路,基本还是采用经典药物的研究方法测定血液中药物的总浓度.纳米药物药动学研究方法的瓶颈是生物样品的前处理技术,大部分研究采用有机溶剂萃取、沉淀蛋白或固相萃取技术等进行样品前处理后只能测定血药总浓度.近年来发展的平衡透析法、超滤离心法、超速离心法和凝胶过滤法等能够测定游离药物浓度,但只能用于体外分析.固相微萃取(SPME)和微透析(MD)技术可以在线取样测定游离浓度,但还需要进一步发展合适的探针和相应的校正定量方法,本文对现有纳米药物药动学研究方法进展进行了评述,并对新的研究方向进行了展望. Development of nanomaterials-based drug delivery systems(NDDS or nanomedicines)represents a booming area of research on both academic and industrial sides globally.However,the regulatory policies for nanomedicines in terms of their safety risks(toxicity)have not been well-established yet.The main challenge is the lack of efficient analytical methodologies to accurately assess the concentrations of nanomedicines in various forms(free,bound or encapsulated)in complicated biological systems.At present,the research ideas of pharmacokinetics of nanomedicines are mainly to determine the total concentration of nanomedicines in blood as usual.The bottleneck of the pharmacokinetic study of nanomedicines is the pretreatment method of biological samples.Most studies use organic solvent extraction,precipitated protein or solid phase extraction technology as pretreatment method to determine the total concentration of nanomedicines in blood samples.Such pretreatment methods as equilibrium dialysis,ultrafiltration centrifugation,ultracentrifugation and gel filtration,etc.developed in recent years can determine the concentration of free medicines,but can only be used for in vitro analysis.Solid phase micro-extraction(SPME)and microdialysis(MD)techniques can be used for on-line sampling and determination of free concentration,but appropriate probes and corresponding calibration methods need to be further developed.In this paper,the present methods of pharmacokinetics of nanomedicines are reviewed and the new research directions are prospected.
