摘要
目的探讨铜绿假单胞菌-甘露糖敏感血凝菌毛株(PA-MSHA)和普鲁卡因(Procaine)诱导膀胱癌细胞凋亡机制。方法将PA-MSHA和普鲁卡因分别与T24、5637膀胱癌细胞共孵育,同时设立磷酸盐缓冲液(PBS)阴性对照组,利用分光光度法及反转录-聚合酶链反应(RT-PCR)法分别检测半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-3/9蛋白和表皮生长因子受体(EGFR)/基质金属蛋白酶(MMP)-9/可溶性Fas受体配体(sFasL) mRNA表达。采用χ^(2)检验或Fisher’s四格表精确检验。结果普鲁卡因和PA-MSHA处理后的膀胱癌细胞出现细胞凋亡的特征性形态学变化;普鲁卡因和PA-MSHA能够活化T24细胞中Caspase最主要的效应蛋白——Caspase-3及Caspase-9;PA-MSHA组MMP-9 mRNA表达量低于对照组50%,sFASL蛋白有20%~35%的升高,EGFR受体升高15%;普鲁卡因组MMP-9 mRNA表达量低于对照组20%~30%,sFASL蛋白mRNA表达量升高15%,差异有统计学意义(P<0.05)。结论 PA-MSHA和普鲁卡因诱导T24及5637膀胱癌细胞的凋亡,与活化Caspase -3/9级联酶蛋白、上调sFASL表达相关,与降低MMP-9金属基质蛋白酶表达有关。
Objective To explore the mechanism of pseudomonas aeruginosa-mannose-sensitive hemagglutinating fimbriae strain(PA-MSHA)and procaine inducing bladder cancer cell apoptosis.Methods PA-MSHA and procaine were incubated with T24 and 5637 bladder cancer cells,respectively,and the expression of cysteinyl aspartate-specific protease(Caspase)-3/9 protein and epidermal growth factor receptor(EGFR)/matrix metalloproteinase(MMP)-9/soluble Fas receptor ligand(sFasL)mRNA was detected by spectrophotometry and reverse transcriptase-polymerase chain reaction(RT-PCR).At the same time,a phosphate buffer saline(PBS)negative control group was established.Usingχ^(2) test or Fisher′s four-grid table exact test,P<0.05 was considered statistically significant difference.Results Bladder cancer cells treated with procaine and PA-MSHA showed characteristic morphological changes of apoptosis.Procaine and PA-MSHA could activate the most important effector proteins of Caspase in T24 cells—Caspase-3 and Caspase-9;PA-MSHA could activate Caspase-9 protein in 5637 cells,but procaine could not activate Caspase-3 and Caspase-9 proteins in 5637 cells.The expression of MMP-9 mRNA drops to about 50%of the control group after PA-MSHA,and sFASL receptor have increased in 20%-35%,EGFR receptor increased by 15%;MMP-9 mRNA expression decreased to about 20%-30%of the control group after procaine,sFASL receptor mRNA expression increased by about 15%.The difference was statistically significant(P<0.05).Conclusion PA-MSHA and procaine induce apoptosis of T24 and 5637 bladder cancer cells,which may be related to the activation of Caspase-3/9 cascade protein,up-regulation of the expression of sFASL,and down-regulation of the expression of MMP-9 metal matrix protease.
