摘要
目的:探讨海带多糖(LJP)对放射(Rad)诱导的小鼠下颌下腺炎症反应的抑制作用,并初步探讨其可能机制。方法:96只雄性昆明小鼠随机分为对照(Ctrl)组、LJP组、Rad组和LJP+Rad组,每组24只。在Rad处理前1周连续给药,采用[^(60)Co]γ射线创建放射性下颌下腺损伤模型并观察小鼠Rad处理后1 d、3 d、7 d和14 d的唾液量变化;采用苏木精-伊红(HE)染色观察下颌下腺组织形态学改变;免疫组织化学法检测下颌下腺组织中核因子κB抑制蛋白α(IκBα)、c-Jun氨基末端激酶(JNK)、磷酸化JNK(p-JNK)、细胞间黏附分子1(ICAM-1)和单核细胞趋化蛋白1(MCP-1)的蛋白表达;ELISA法检测血清中肿瘤坏死因子α(TNF-α)、MCP-1及巨噬细胞炎症蛋白2(MIP-2)的表达;免疫荧光法检测下颌下腺核因子κB(NF-κB)p65亚基和JNK的定位。结果:与Ctrl组相比,Rad组小鼠唾液量显著降低(P<0.05),镜下可见大量炎症细胞浸润,且NF-κB和JNK信号通路相关炎症因子和蛋白表达显著增强(P<0.05);而LJP干预可以减轻小鼠下颌下腺炎症,并显著抑制NF-κB和JNK信号通路相关蛋白的表达。结论:LJP可以减轻Rad诱导的小鼠下颌下腺炎症反应,其机制可能与抑制NF-κB和JNK信号通路有关。
AIM:To investigate the effect of Laminaria japonica polysaccharides(LJP)on radiation(Rad)-induced submandibular gland inflammation and to study the possible mechanism.METHODS:A total of 96 Kunming mice were randomly divided into control(Ctrl)group,LJP group,Rad group and LJP+Rad group,with 24 mice in each group.The LJP was administered continuously one week before radiation.[^(60)Co]γ-ray irradiation at 15 Gy was used to established the salivary gland injury model.The changes of salivary volume at 1 d,3 d,7 d and 14 d after irradiation were measured.The protein levels of inhibitor of nuclear factor-κBα(IκBα),c-Jun N-terminal kinase(JNK),phosphorylated JNK(p-JNK),intercellular adhesion molecule-1(ICAM-1)and macrophage chemoattractant protein-1(MCP-1)were determined by immunohistochemistry.The serum levels of tumor necrosis factor-α(TNF-α),MCP-1 and macraphage inflammathory protein-2(MIP-2)were measured by ELISA.The morphological changes of submandibular gland were observed by HE staining,and the localization of nuclear factor-κB(NF-κB)p65 subunit and JNK in the submandibular gland was examined by immunofluorescence.RESULTS:The volume of saliva in Rad group was significantly decreased as compared with control group(P<0.05),and a large number of inflammatory cells were observed.The levels of inflammatory factors and proteins related to NF-κB and JNK signaling pathways were significantly increased in Rad group(P<0.05).After intervention with LJP,the salivary gland inflammation of the mice was alleviated,and the expression of NF-κB and JNK signaling pathway-related proteins was significantly inhibited(P<0.05).CONCLUSION:The LJP attenuate radiation-induced inflammatory injury of submandibular gland in mice,and the mechanism may be related to the inhibition of NF-κB and JNK signaling pathways.
