摘要
目的:优化红豆树叶总黄酮提取工艺并研究其抗氧化性。方法:采用超声辅助法分别考察液料比、乙醇浓度和超声时间对红豆树叶总黄酮提取率的影响。以单因素试验为依据,进行响应面设计试验及数据分析,进而对其工艺进行优化设计,同时进行总黄酮的抗氧化性试验。结果:各因素对提取率的影响由大到小顺序为乙醇浓度、液料比和超声时间;且最佳提取工艺为乙醇浓度69.45%,液料比20.15∶1(mL/g),超声时间90.17 min。在此工艺下,总黄酮提取率为23.85%,通过验证试验发现,试验提取率为23.87%,与预测值相对标准偏差为0.06%;同时红豆树叶总黄酮对DPPH自由基和ABTS自由基表现出较好的清除效果。结论:优化后的红豆树叶总黄酮提取工艺稳定可行,并且其具有较强的抗氧化能力。
Objective:Optimization of the extraction technology of total flavonoids from leaves of Ormosia hosiei and study on its antioxidant activity.Methods:The effects of liquid to solid ratio,ethanol concen⁃tration and ultrasonic time on the extraction rates of total flavonoids from leaves of O.hosiei were investi⁃gated by ultrasonic-assisted extraction.Based on the results of single-factor test,response surface design and data analysis were carried out to further optimize the ultrasonic-assisted extraction.At the same time,the antioxidant activity of total flavonoids was tested.Results:The influence of factors on flavo⁃noids extraction rate was in turn ethanol concentration,liquid to solid ratio,the ultrasonic time.The opti⁃mal extraction process conditions were liquid to solid ratio20.15∶1(mL/g),ethanol concentration 69.45%and ultrasonic time 90.17 min.Under the above conditions,the extraction of total flavonoids was 23.85%.The experimental results showed that the extraction of total flavonoids was 23.87%,which was close to the predicted value and the relative standard deviation between the two was 0.06%.Meanwhile,finding the total flavonoids from leaves of O.hosiei had good scavenging effects on DPPH free radicals and ABTS free radicals.Conclusion:The optimized extraction process of total flavonoids from the leaves of O.hosiei was stable and workable,and it has a strong antioxidant ability.
