摘要
为合理开发和科学利用青檀植物内生真菌资源,以发酵粗提液的二苯代苦味肼基自由基(DPPH)清除率作为检测指标,筛选青檀内生真菌高抗氧化活性菌株,通过单因素和正交试验进行培养条件的优化研究。结果表明:在供试的30株青檀内生真菌中具有抗氧化活性的菌株有3株,占10%。高抗氧化活性菌株的最适发酵培养条件是:葡萄糖20 g/L、(NH_(4))_(2)SO_(4)2 g/L、K_(2)HPO_(4)3.2 g/L、CuSO_(4)6.0×10^(-3) g/L、MgSO_(4)0.2 g/L、最适装液量80 mL/250 mL、pH值7.0、温度27℃、培养时间8 d。
In order to rationally develop and utilize the endophytic fungi resources of Pteroceltis tatarinowii,DPPH clearance rate was used as an indicator to screen high oxidation resistant strains and optimized culture by single factor and orthogonal experiments.The results showed that 30 strains of endophytic fungi were selected for determination of antioxidant activity,and 3 strains with antioxidant activity accounted for 10%.The optimal fermentation conditions were:glucose 20 g/L,(NH_(4))_(2)SO_(4)2 g/L,K_(2)HPO_(4)3.2 g/L,CuSO_(4)6×10^(-3) g/L,MgSO_(4)0.2 g/L,optimum liquid volume 80 mL/250 mL,pH 7.0,temperature 27℃,culture time 8 d.
