摘要
具有血小板结合蛋白基序的解聚蛋白样金属蛋白酶-12(a disintegrin and metalloproteinase with thrombospondin motifs-12,ADAMTS-12)是一种多结构域组成的金属蛋白酶,不同于以降解细胞外基质闻名的其他ADAMTS家族成员,越来越多的报道发现,ADAMTS-12在肿瘤中起到抑癌和促癌的双向作用。ADAMTS-12除发挥蛋白水解酶功能外,其裂解的活性蛋白产物常参与到肿瘤细胞之间,以及细胞与基质的相互作用中,是肿瘤发生发展的关键调节因子。该文综述了ADAMTS-12的结构和功能,并重点回顾了ADAMTS-12在肿瘤发生发展中较为复杂的促癌和抑癌双向作用及机制,以为肿瘤的靶向治疗提供新的思路。
ADAMTS-12(a disintegrin and metalloproteinase with thrombospondin motif-12)is a multi-domain metalloproteinase,which is different from other members of the family known for degradation of extracellular matrix.More and more reports have found that ADAMTS-12 plays a dual role in tumor inhibition and tumor promotion.In addition to playing a proteolytic enzyme function,the split products of ADAMTS-12 protein are often involved in cell-to-cell and cell-to-matrix interactions,and is the key regulation factor in the tumor development.This review summarizes the structure,functions and mechanism of ADAMTS12 in tumor inhibition and tumor promoting,so as to provide a new idea for the targeted therapy of tumor.
