马蹄金花粉活力检测的对比试验分析 被引量:3

摘要 本试验以前期研究筛选出的坪用价值、抗逆性均表现较好,不同材料为主要研究对象,分别从花粉活力与柱头可授性等方面,探讨造成其开花特性差异。主要结果如下:比较四种不同方法测定野生马蹄金花粉生活力效果显示:碘-碘化钾染色法、TTC法染色效果不佳,不适宜野生马蹄金花粉活力测定;联苯胺-甲萘酚染色法能测定花粉生活力,但测定值偏高;离体萌发法测定野生马蹄金花粉活力,花粉管生长良好,且萌发率基本稳定,便于快速直观统计花粉生活力,适宜野生马蹄金花粉生活力检测,其较适宜的培养基为蔗糖10%+硼酸(HBO_(3))0.005%+硝酸钙[Ca(NO_(3))_(2)·4H_(2)O]0.015%+硫酸镁(MgSO_(4)·7H_(2)O)0.001%+硝酸钾(KNO_(3))0.005%+0.5%琼脂。利用离体萌发法测定SD200303和GD200503花粉离体萌发率分别为54.96%和2.43%,花粉寿命分别为5h和1h。 The objective of the study was find the reasons of low fecundity between the native Dichondra repens from soral biology,pollen viability,stigma receptivity,the main results are follows:The pollen viability of wild Dichondra repens SD200303 and GD200503 was detected by 4 diffeient methods respectively.The comparwason of different methods indicated that methods of the I2-KI、TTC were not suitable to evaluate wild dichondra repens pollen activity;4,4'-diaminobiphenyl-1-Naphthol method could detect the pollen viability,but the measured value was higher;the method of germination in vitro was the fittest,which pollen tube grew well and pollen viability was stable that was convenient for determination.The optimum culture condition was:10% Sucrose+0.005%HBO_(3)+0.015%[Ca(NO_(3))_(2)·4H_(2)O]+0.001% MgSO_(4)·7H_(2)O+0.005% KNO_(3)+0.5% Agar.The pollen germination rate was dectected by pollen germination in vitro showed that the pollen germination rate of SD200303 and GD200503 was 54.96%and 2.43%,pollen life span was 5h and 1h,respectively.
