柠檬精油的GC-MS分析及其抗炎、抗菌活性研究 被引量:6

GC-MS analysis of lemon essential oil and its anti-inflammatory and antibacterial activities

摘要 为研究柠檬精油的组分、体外抗炎及其抑菌活性,利用GC-MS对3种精油进行分析,采用峰面积归一化法计算相对含量;通过LPS建立RAW264.7炎症模型,凭借ELISA法评价抗炎效果;使用牛津杯法测定其对金黄色葡萄球菌、大肠埃希菌及白色念球菌的抑菌效果.3种精油通过GCMS共检出26种挥发性成分其中14种为共有成分,萜烯类化合物是最主要的成分且柠檬烯的含量最高(34.37%~50.87%);3种精油在一定程度上降低由LPS诱导增加的TNF-α(P<0.05),但3种精油不同质量浓度时抑制效果均未表现良好的剂量依赖关系;3种精油对3种受试菌均有抑制作用,甜柠檬精油抑菌效果为中敏,绿柠檬精油抑制大肠埃希菌为高敏,其余为极敏,黄柠檬精油的抑菌作用最佳,抑菌效果均为极敏.因柠檬精油具有多种生物活性成分,故具有一定的体外抗炎作用和良好的抑菌活性. In order to study the composition of lemon essential oil and its anti-inflammatory and antibacterial activities in vitro,GC-MS was used to analyze the three essential oils,and the relative content was calculated by peak area normalization method;the RAW 264.7 inflammation model was established by LPS,and detected by ELISA method to evaluate the anti-inflammatory effect;the Oxford Cup method was used to determine the antibacterial activity against Staphylococcus aureus,Escherichia coli and Candida albicans.It turned out that 26 kinds of volatile components were detected by GC-MS in the three essential oils of which 14 kinds were common components,terpenes were the most important component and the content of limonene was the highest(34.37%~50.87%);for three essential oils,the increase of TNF-αinduced by LPS(P<0.05)in a certain degree,but the inhibitory effects of the three essential oils at different mass concentrations did not show a good dose-dependent relationship;the three essential oils had inhibitory effects on the three tested bacteria The antibacterial effect of sweet lemon essential oil was medium sensitive,green lemon essential oil was highly sensitive to Escherichia coli,and the rest was extremely sensitive.Yellow lemon essential oil had the best antibacterial effect and the antibacterial effect was extremely sensitive.Because lemon essential oil has a variety of biologically active ingredients,it has a certain in vitro anti-inflammatory effect and good antibacterial activity.
