针刀干预对膝骨关节炎兔股二头肌细胞凋亡的影响 被引量:5

Effects of Acupotomy Intervention on Apoptosis of Biceps Femoris Cells in KOA Rabbits

摘要 目的:探讨针刀松解股二头肌对膝骨关节炎(KOA)兔肌细胞凋亡的影响。方法:6月龄新西兰兔48只随机分为正常组(N)、模型组(M)、电针组(EA)和针刀组(A),KOA模型采用改良后维德曼左后肢伸直位固定制动法制动8周,于造模成功1周后分别对A组、EA组进行针刀和电针干预3周。干预结束1周后取股二头肌进行HE染色观察、TUNEL检测阳性凋亡细胞核数和Western blot检测Bax、Caspase-3蛋白表达水平。结果:A组细胞核和间质组织的水肿、炎性肌细胞与M组相比显著改善,改善程度优于EA组。M组兔股二头肌阳性细胞核数较N组明显升高(P<0.01);EA组较N组明显升高(P<0.01);EA组较M组有所降低(P<0.05);A组较N组明显升高(P<0.01);A组较M组明显降低(P<0.01);A组较EA组明显降低(P<0.05)。M组兔股二头肌Caspase-3蛋白表达水平较N组明显升高(P<0.01);EA组与N组比较差异无统计学意义(P>0.05);A组与N组比较差异无统计学意义(P>0.05);EA组较M组有所降低(P<0.05);A组较M组明显降低(P<0.01);A组与EA组比较差异无统计学意义(P>0.05)。结论:针刀干预对KOA股二头肌细胞的凋亡有良好的调节作用,为针刀“调筋治骨”提供了新的理论依据。 Objective:To study the effect of acupotomy intervention on the apoptosis of biceps femoris cells in the rabbits with knee osteoarthritis(KOA).Methods:48 New Zealand rabbits aged 6 months were randomly divided into the normal group,the model group,the electro-acupuncture(EA)group and the acupotomy group.The KOA model was prepared by modified Wiedemans left hind limb extension fixation braking method.After one week of successful modeling,acupotomy and electro-acupuncture intervention were performed in the acupotomy group and the EA group for three weeks,respectively.One week after the end of the intervention,biceps femoris muscle was collected for HE staining observation,TUNEL detection and Western blot were used to detect the protein expressions of Bax and caspase-3.Results:The edema of nucleus and interstitial tissues and inflammatory muscle cells in the acupotomy group were significantly improved compared to those in the model group,and the improvements in the acupotomy group were better than those in the EA group.In terms of the nucleus number of positive biceps femoris,it was significantly higher in the model group than that in the normal group(P<0.01);it was significantly higher in the EA group than that in the normal group(P<0.01),and was significantly lower than that in the model group(P<0.05);it was significantly higher in the acupotomy group than that in the normal group(P<0.01),and was lower than that in the model group(P<0.01);it was significantly lower in the acupotomy group than that in the EA group(P<0.05).In terms of the protein expression of Caspase-3 of biceps femoris,it was significantly increased in the model group than that in the normal group(P<0.01);there was no significant difference between the EA group and the normal group(P>0.05);there was also no statistical difference between the acupotomy group and the normal group(P>0.05);it was lower in the EA group than that in the model group(P<0.05);it was also significantly lower in the acupotomy group than that in the model group(P<0.01);there was no significant difference between the acupotomy group and the EA group(P>0.05).Conclusion:Acupotomy intervention had a good regulatory effect on the apoptosis of biceps femoris cells in the treatment of KOA,and it provided a new theoretical basis for the‘adjustment of tendons and treatment of bone’of acupotomy.
