饲料锌对长江鲟幼鱼肠道微生物组成的影响 被引量:4

Effects of dietary zinc on the intestinal microbiota composition of juvenile Acipenser dabryanus

摘要 为了解锌对长江鲟(Acipenser dabryanus)类肠道菌群的多样性及群落结构组成的影响,本研究配制了14.77,57.71和364.5 mg/kg的3种不同锌水平的饲料,养殖长江鲟幼鱼8周,采集了后肠粪便,测定了后肠粪便的肠道α-多样性、群落结构组成与OTU数量。结果显示:随饲料锌水平的升高,基于丰度的覆盖估计值(ACE指数)、Observed species指数、Chao1丰富度估计指数(Chao1指数)出现了下降的趋势,364.5 mg/kg组最低,364.5 mg/kg组与14.77 mg/kg组之间差异显著,与57.71mg/kg组之间无显著差异。香浓多样性指数、辛普森多样性指数与普系α-多样性(Phylogenetic diversity,PD-whole tree)值也表现出随饲料锌水平的升高而下降的趋势,且三种指数均在364.5 mg/kg时最低,但各组之间无显著性差异。在科水平上,梭杆菌科、肠杆菌科与梭菌科类的总和分别占前10种科的80.82%、92.89%、93.37%,表明在不同锌饲料处理水平下,上述3个菌科在鱼类肠道菌群的组成中占据优势。在属水平上,鲸杆菌属(Cetobacterium)、梭菌属类(unidentified_Clostridiales)、邻单胞菌属(Plesiomonas)与柠檬酸杆菌属(Citrobacter)总和分别占前10种属的总和百分比分别达78.32%、81.56%、89.28%,表明这4种细菌属是优势属。三个处理组的总OTU数随饲料锌水平的升高而出现了下降的趋势,三个处理组的OTU数分别为1160、790、654。上述结果表明,饲料锌含量对长江鲟幼鱼肠道的α-多样性、群落结构组成及OTU数量产生了影响,且饲料锌含量过高时会影响长江鲟幼鱼肠道的α-多样性、群落结构丰度及OTU数。 This study formulated three diets with different zinc levels(14.77,57.71 and 364.5 mg/kg)to breed juvenile Acipenser dabryanus for 8 weeks,hindgut feces were collected and the intestinalα-diversity,community composition and the numbers of OTU in the hindgut were measured.The results indicated that the abundance-based coverage estimator(ACE index),observed species index and Chao1 index showed a downward trend with the increasing of dietary zinc contents.The three indexes reached the lowest value in fish fed supplemented with 364.5 mg/kg zinc diets,which was significant different between the 14.77 mg/kg group and 364.5 mg/kg group.However,no significant difference was observed between the 57.71 mg/kg zinc group and 364.5 mg/kg zinc group.The Shannon index,Simpson index and phylogenetic diversity-whole tree(PD-whole tree)value also showed a downward trend with the increasing of dietary zinc contents.The three indexes reached the lowest value in fish fed supplemented with 364.5mg/kg zinc diets.No significant difference was observed among the three groups.At the family level,the sum of Fusobacteriaceae,Enterobacteriaceae,and unidentified_Clostridiales accounted for 80.82%,92.89%and 93.37%in the top 10 families,respectively,which indicated that the three families had advantage in the different zinc contents diets.At the genus level,the sum of Cetobacterium,unidentified_Clostridiales,Plesiomonas,and Citrobacter accounted for 78.32%,81.56%,and 89.28%in the top 10 genera,respectively,which indicated that the four genus were advantage genera.The total OTU number of the three treatments showed downward trends with the increasing of dietary zinc contents.The numbers of OTU in the three treatments were 1160,790 and 654,respectively.The above results suggested that intestinalα-diversity,community structure and OTU number of juvenile Acipenser dabryanus influenced by the dietary treatments,and too higher zinc in the dietary would reduced the intestinalα-diversity,the relative abundance of community and the numbers of OTU.
