摘要
目的观察紫归长皮软膏治疗大鼠难愈性创面的效果。方法用SD大鼠制备难愈性创面模型,随机分为正常空白组(n=8)、模型空白组(n=8)、紫归长皮软膏组(n=8)、贝复济组(n=8),给予创面不同的治疗方案。通过各组肉芽组织HE染色及神经营养因子(Nerve growth factor,NGF)的免疫组化检测,观察紫归长皮软膏对大鼠难愈性创面愈合的促进作用及神经生长因子的分泌情况。结果正常空白组及模型空白组于创伤后第7天,均有单核细胞渗出,伴有成纤维细胞和毛细血管的增生,第10天皮下成纤维细胞生长较活跃,数量多,其毛细血管胚芽形成数量仍较少;中药紫归长皮软膏组及贝复济组于创伤后第7天,基本无单核细胞渗出,伴有大量成纤维细胞和毛细血管增生,第10天皮下成纤维细胞生长活跃,数量多,毛细血管胚芽数量显著增多;紫归长皮软膏组优于贝复济组。各组均有NGF表达,在创伤后第7天及第10天,与模型空白组比较,紫归长皮软膏组及贝复济组NGF表达均更高(P<0.05),且紫归长皮软膏组高于贝复济组(P<0.05)。结论紫归长皮软膏能明显促进成纤维细胞和毛细血管的增生,并能显著促进神经营养因子的分泌,进而促进大鼠慢性难愈性创面愈合。
Objective To observe the effect of Zigui Zhangpi Paste on the refractory wound of rats.Methods Refractory wound model of SD rats was established.The rats were randomly divided into 4 groups:normal blank group(n=8),modal blank group(n=8),Zigui Zhangpi Paste group(n=8)and bFGF-torita group(n=8).They were given different schemes of wound treatment.Through HE staining of granulation tissue and immunohistochemistry of nerve growth factor(NGF)in each group,the promoting effect of Zigui Zhangpi Paste on the refractory wound of rats and the secretion of NGF were observed.Results In the normal blank group and the model blank group,on the 7th day after trauma,there was monocyte exudation,accompanied by proliferation of fibroblasts and capillaries.On the 10th day after trauma,the growth of subcutaneous fibroblasts was more active,and the number of capillary germ formation was still less.In the Zigui Zhangpi Paste group and the bFGF-torita group,on the 7th day after trauma,there was no monocyte exudation,with a large number of fibroblasts and capillaries proliferation.On the 10th day after trauma,the number of subcutaneous fibroblasts was more active,and the number of capillary germ was significantly increased.The Zigui Zhangpi Paste group was better than the bFGF-torita group.NGF was expressed in all groups.On the 7th and 10th day after trauma,compared with the model blank group,NGF expression was higher in the Zigui Zhangpi Paste group and the bFGF-torita group(P<0.05),and higher in the Zigui Zhangpi Paste group than in the bFGF-torita group(P<0.05).Conclusion Zigui Zhangpi Paste can promote the proliferation of fibroblasts and capillaries,and the secretion of neurotrophic factors,thus promoting the healing of refractory wound in rats.
