摘要
目的建立升降茶高效液相指纹图谱共有模式,并测定其芦荟大黄素、姜黄素、大黄酸、大黄素、大黄酚5种成分的含量。方法采用HPLC法,色谱柱为Phenomenex Lune C_(18)(5μm,4.6 mm×250 mm),流动相为甲醇-0.1%磷酸溶液(梯度洗脱),流速为1.0 mL·min^(-1),检测波长254 nm,柱温为25℃。采用中药指纹图谱软件(2012A版)、SPSS 22.0软件对10批样品进行相似度分析和聚类分析。以姜黄素为内标,采用一测多评法(QAMS)测定升降茶中5种成分含量。结果10批样品指纹图谱共标定出12个共有峰,相似度均在0.98以上;芦荟大黄素、姜黄素、大黄酸、大黄素、大黄酚5种成分在各自范围内与峰面积线性关系良好(r≥0.9993),平均加样回收率分别为100.2%(RSD=1.1%)、99.8%(RSD=0.30%)、99.6%(RSD=0.53%)、99.9%(RSD=0.85%)、100.3%(RSD=1.2%),QAMS所得结果与外标法无明显差异(RSD<2%)。结论该研究建立的指纹图谱和QAMS含量测定法稳定可控,简单易行,可用于升降茶的质量控制。
Objective To establish the HPLC fingerprint common model of Shengjiang tea,and to determine the contents of aloe emodin,curcumin,rhein,emodin and chrysophanol.Methods HPLC was used,the column was Phenomenex Lune C18(5μm,4.6 mm×250 mm),and the mobile phase was methanol-0.1%phosphoric acid solution(gradient elution).A gradient elution was used at 1.0 mL·min^(-1),the detection wavelength was 254 nm,while the column temperature was 25℃.The similarity evaluation software(2012A)and SPSS 22.0 were used to analyze the similarity and cluster of 10 batches of samples.Curcumin was used as the internal standard to determine the content of 5 components by quantitative analysis of multi-components by single-marker(QAMS).Results The similarity of 10 batches of Shengjing tea was above 0.98,the linearity of 5 components was good(r≥0.9993),the average recovery was 100.2%(RSD=1.1%),99.8%(RSD=0.30%),99.6%(RSD=0.53%),99.9%(RSD=0.85%),and 100.3%(RSD=1.2%).There was no significant difference between the results of QAMS and external standard method(RSD<2%).Conclusion The fingerprints and QAMS method are stable and controllable,which can be used for the quality control of Shengjiang tea.
