摘要
Wnt/β-连环蛋白(Wnt/β-catenin)信号通路已经被证明与人体多系统疾病的发生和发展有关。近年来,寻找该信号通路的特异性调控药物,一直是研究热点。但有研究认为关键因子β-连环蛋白(β-catenin)对某些疾病是非成药靶点,不能与小分子化合物良好结合;且由于该通路调控的基因众多等原因,目前针对该通路的新药研发复杂而困难,国际上仍没有批准上市的靶向该通路的治疗药物。中药特别是中药复方具有长期的临床用药经验和多靶向性等特点,中药复方调控该通路治疗多系统疾病的研究近年来也在广泛开展。本文就近年来中药复方对该信号通路的调控研究进行文献综述,以期筛选出部分经典方剂或经验方,为下一步本课题组采用基于高通量基因测序技术的高通量基因筛查技术(HTS2)研究和验证这些复方对该通路的调控机制和建立该通路的中药-细胞-基因数据库奠定基础。
Wnt/β-catenin signal pathway has been proved to be relate to the occurrence and development of multiple system diseases. Many researches always focus on discovering some specific agents that can regulate this signal pathway in recent years. However, some researches have discovered that as the key gene of this signal pathway, β-catenin is an undruggable target without the ability to combine with small molecule compound effectively. Many other genes can be regulated by this signal pathway, which makes new drug development acting on this signal pathway difficult and complicated, there is still no granted drug for clinical use. Chinese herbal medicine, especially the compound Chinese medicine has the characters of long time clinical use and multiple effective targets. The researches of compound Chinese medicine to control this signal pathway have been in extensive development in recent years. We made a review about the researches of compound Chinese medicine on the regulation of this signal pathway in order to find some classical prescriptions or empirical formulas. Then we will use the "High Throughput Sequencing based High Throughput Screening"( HTS 2) technology to study and testify controlling effects of these prescriptions and formulas on Wnt/β-catenin signal pathway and establish Chinese herbal medicine-cell-gene database.
