马铃薯StDRO2基因的生物信息学分析 被引量:2

摘要 【目的】根系构型对植物生长发育起到重要作用,DEEPER ROOTING1(DRO1)基因参与调控植物根生长角度。但是马铃薯DRO1同源基因的生物学信息还不清楚。【方法】本研究利用生物信息学软件分析了马铃薯中StDRO2蛋白的理化性质、亚细胞定位、保守结构域、系统发育树及蛋白质结构。【结果】发现马铃薯StDRO2基因编码亲水性不稳定蛋白,定位在细胞核,氨基酸序列具有IGT基因家族典型结构域。蛋白质二级结构预测显示StDRO2蛋白质均具有无规则卷曲、α-螺旋、β-转角和延伸链结构。通过实时定量PCR分析,马铃薯幼苗叶片中StDRO2的表达量显著高于根和茎杆。生长素处理后,根中StDRO2基因的表达量显著降低,说明该基因的表达受到生长素的负调控。【结论】StDRO2属于IGT基因家族中的一员,可能参与调控马铃薯根系构型。马铃薯StDRO2基因的生物信息学及表达水平检测为今后分析马铃薯根系发育提供了理论基础。 【Objective】Root system architecture plays an important role in plant development,and function of DEEPER ROOTING 1(DRO1)gene involves in regulating plant root growth angle.However,the biological information of potato DRO1 homologous gene remains unclear.【Method】In the present study,bioinformatics softwares were used to analyze the physical and chemical properties,subcellular localization,conserved domain,phylogenetic tree and protein structure of StDRO2 protein in potato.【Result】Results showed that the potato StDRO2 gene encoded a hydrophilic and unstable protein located in the nucleus and the amino acid sequence contained typical domain of IGT gene family.The protein secondary structure showed that StDRO2 protein had random curl,α-helix,β-turn and extended chain structure.Real-time quantitative PCR analysis showed that the expression level of StDRO2 in potato seedling leaves was significantly higher than that in roots and stems.Auxin treatment down regulated the expression level of StDRO2 gene in roots.It indicated that the expression of StDRO2 gene was negatively regulated by auxin.【Conclusion】StDRO2 was a member of IGT gene family and involved in regulating potato root system architecture.The bioinformatic and expression assay of potato StDRO2 protein provided us theoretical basis to analyze potato root development in future research.
