摘要 目的观察五味子酯甲(Schisantherin A,SCA)对人肝癌细胞(HepG_(2))增殖和凋亡的影响.方法将体外培养的HepG_(2)细胞分为空白对照组(CON)和SCA 25、50、100μmol/L 3个浓度组.给药48 h后,应用MTT法检测细胞存活率;应用Western blot法检测HepG_(2)细胞Bcl-2、Bax和Caspase-3蛋白的表达水平;Hoechst染色观察上述各组细胞凋亡情况.结果与CON组比较,SCA各组HepG_(2)细胞存活率均显著降低(P<0.01);Bax及Caspase-3蛋白的表达水平显著增加(P<0.01),Bcl-2蛋白及Bcl-2/Bax比值表达显著降低(P<0.05或P<0.01);Hoechst染色显示HepG_(2)细胞出现明显的核浓缩及核碎裂形态.结论SCA能够抑制HepG_(2)细胞增殖并促进其凋亡,该作用可为开发利用SCA的相关药物提供理论依据. Objective To observe the effects of Schisantherin A(SCA)on proliferation and apoptosis of human hepatoma cells(HepG_(2)).Method HepG_(2) cell line was cultured in vitro and divided into four groups:control group(CON)and three SCA groups(25μmol/L,50μmol/L and 100μmol/L).After 48 hours of the treatment,the cell survival rate was detected by MTT assay;The expressions of Bcl-2,Bax and Caspase-3 protein in CON and SCA were detected by Western blot;The apoptosis in each group was observed by Hoechst staining.Results In comparison to the control group,the cell survival rates in SCA groups decreased significantly(P<0.01);The protein expression levels of Bax and Caspase-3 significantly increased(P<0.01),and the Bcl-2 protein and Bcl-2/Bax ratio significantly decreased(P<0.05 or P<0.01).Hoechst staining showed significant nuclear concentration and fragmentation.Conclusion SCA can inhibit the proliferation of HepG_(2) cells and promote their apoptosis,which provides a theoretical basis for the development of SCA.
