基于Th1/Th2失衡及炎症反应探讨封髓丹加味对痤疮的影响 被引量:7

摘要 目的:探究封髓丹加味对痤疮大鼠炎症反应的影响及其机制。方法:将40只雄性SD大鼠随机分为对照组、模型组、封髓丹加味低、中、高剂量治疗组5组各8只,于大鼠耳廓皮下注射痤疮丙酸杆菌(P.ances)建立痤疮大鼠模型,治疗组分别以含生药1.75 g/mL、3.5 g/mL及7 g/mL的封髓丹加味进行灌胃处理,连续处理7 d;观察大鼠耳部皮损情况及组织病理变化,计数炎性细胞的数量,采用酶联免疫吸附法(ELISA)检测Th1细胞分泌因子白细胞介素2(interleukin-2,IL-2)、干扰素γ(interferon-γ,IFN-γ)和Th2细胞分泌因子白细胞介素4(interleukin-4,IL-4)及白介素10(interleukin-10,IL-10)的含量,蛋白印迹(Western Blot)检测Toll样受体(the toll-like receptor family,TRLs)/核转录因子κB(nuclear transcription factor-κB,NF-κB)通路相关蛋白的表达。结果:封髓丹加味可有效改善痤疮大鼠耳廓增厚、红肿及丘疹症状,减轻耳廓皮损组织中炎性浸润、间质出血和水肿程度。与模型组比较,封髓丹加味高剂量显著下调痤疮大鼠皮损评分、皮损组织中炎性细胞数、IL-4、IL-10、TRL2、TRL4及NF-κB磷酸化水平,显著上调IL-2、IFN-γ水平(P<0.05),差异均有统计学意义。结论:封髓丹加味可有效改善痤疮的炎症反应,其作用机制可能与抑制TRL/NF-κB信号通路、纠正Th1/Th2失衡有关。 Objective:To explore the effects of modified Fengsui Pills on inflammatory reactions in acne rats and its mechanism.Methods:40 male SD rats were randomly divided into 5 groups:control group,model group,low-dose,medium-dose and high-dose modified Fengsui Pill treatment group,8 cases in each group.Subcutaneous injection of propionibacterium acnes(P.ances)was conducted at the auricle of rats to establish acne rat models.The treatment groups were given gavage treatment with modified Fengsui Pills containing 1.75 g/mL,3.5 g/mL and 7 g/m crude drugs.They were continuously treated for 7 d.The ear skin damage and histopathological changes were observed.The number of inflammatory cells was statistically analyzed.The contents of Th1 cell secretion factors[interleukin-2(IL-2),interferon-γ(IFN-γ)]and Th2 cell secretion factors[interleukin-4(IL-4),interleukin-10(IL-10)]were detected by enzyme-linked immunosorbent assay(ELISA).Western blot was applied to detect expression of toll-like receptor family/nuclear transcription factor-κB(TRL/NF-κB)pathway related proteins.Results:Modified Fengsui Pill treatment could effectively improve auricle thickening,redness and papule symptoms of acne rats,alleviate inflammatory infiltration,interstitial hemorrhage and edema degree in auricular skin damage tissues.Compared with model group,skin damage score of acne rats,number of inflammatory cells in skin damage tissues,IL-4,IL-10,TRL2,TRL4 and phosphorylation level of NF-κB were significantly down-regulated in high-dose modified Fengsui Pill treatment group,while levels of IL-2 and IFN-γwere significantly up-regulated(P<0.05).Conclusion:Modified Fengsui Pills can improve inflammatory reactions of acne.And its mechanism may be related to inhibiting TRL/NF-κB signaling pathway and correcting Th1/Th2 imbalance.
