摘要
目的观察糖尿病大鼠骨骼肌中musclin、myonectin mRNA的表达及阿托伐他汀钙对其表达的影响。方法选取8周龄SPF级雄性Wistar大鼠50只,随机分为5组:空白对照组、高糖高脂模型组、糖尿病模型组、高糖高脂治疗组及糖尿病治疗组,每组10只。空白对照组大鼠给予普通饲料喂养,高糖高脂模型组及治疗组大鼠采用高糖高脂饲料喂养,糖尿病模型组及治疗组大鼠采用高糖高脂饲料联合小剂量链脲菌素法行2型糖尿病大鼠造模,造模成功后高糖高脂治疗组和糖尿病治疗组大鼠予阿托伐他汀钙8.4 mg/kg灌胃,余3组使用同等剂量0.9%氯化钠注射液灌胃。6周后,观察光镜下各组大鼠骨骼肌形态学变化,并采用实时定量聚酶链式反应检测大鼠骨骼肌musclin、myonectin mRNA表达水平。结果高糖高脂模型组和糖尿病模型组大鼠骨骼肌musclin、myonectin mRNA表达水平均显著高于空白对照组大鼠(P<0.05);高糖高脂治疗组大鼠musclin、myonectin mRNA表达较高糖高脂模型组大鼠明显降低(P<0.05);糖尿病模型组与糖尿病治疗组musclin、myonectin mRNA表达比较差异无统计学意义(P>0.05)。结论高糖高脂状态下大鼠骨骼肌中musclin、myonectin mRNA表达增强,使用阿托伐他汀钙干预后可明显抑制糖尿病前期大鼠musclin、myonectin mRNA表达,但对糖尿病大鼠无明显改善作用。
Objective To observe the mRNA expression of musclin and myonectin in skeletal muscle of diabetic rats and the effect of atorvastatin calcium on their expression.Methods Fifty 8-week-old male Wistar rats were randomly divided into 5 groups:blank control group,high glucose and high fat model group,diabetes model group,high glucose and high fat treatment group and diabetes treatment group,with 10 rats in each group.The rats in the blank control group were fed with normal diet;the rats in the high sugar and high fat model group and treatment group were fed with high sugar and high fat diet;the rats in the diabetes model group and diabetes treatment group were fed with high sugar and high fat diet combined with a small dose of streptozotocin to establish the model of type 2 diabetes mellitus(T2DM).After successful modeling,the rats in the high sugar and high fat treatment group and the diabetes treatment group were given atorvastatin calcium 8.4 mg/kg by intragastric gavage.The other three groups were given the same dose of 0.9%sodium chloride injection by intragastric gavage.After 6 weeks,the morphological changes of skeletal muscle in the three groups were observed under light microscope,and the expression levels of musclin and myonectin were detected by real-time quantitative PCR.Results The mRNA expression levels of musclin and myonectin in high glucose and high fat model group and diabetes model group were significantly higher than those in blank control group(P<0.05);the mRNA expression levels of musclin and myonectin in high glucose and high fat treatment group were significantly lower than those in high glucose and high fat model group(P<0.05).There was no significant difference in the mRNA expression levels of musclin and myonectin in diabetes model group and diabetes treatment group(P>0.05).Conclusion The mRNA expression of musclin and myonectin in skeletal muscle of rats under high glucose and high fat condition is enhanced.Atorvastatin calcium intervention can significantly inhibit the mRNA expression of musclin and myonectin in pre-diabetic rats,but has no significant effect on diabetic rats.
