摘要
本试验以火力楠种子园内5个无性系花粉为材料,测定了不同无性系间的花粉生活力,研究了花粉贮藏的适宜温度及花粉离体萌发的最佳培养基。结果表明,(1)各无性系间花粉活力存在差异,无性系序号排序由大到小为42>14>51>48>7;(2)各无性系花粉在不同贮藏温度下活力随时间的增长呈逐渐下降趋势,常温条件下火力楠花粉活力下降较快,而-20℃贮藏条件下活力保存最好,在-20℃贮藏14 d后42号无性系花粉活力仍可达到39.28%;(3)火力楠5个无性系花粉的最佳培养基配方为100 g/L蔗糖+50 mg/L硼酸+1%琼脂粉。
Pollens from five different clones in Michelia macclurei clonal seed orchard were used as materials to measure pollen viability and study the suitable storage temperature and the best medium for pollen germination in vitro. Results showed that,(1) Pollen viability was different among clones,ranking as follows: 42>14>51>48>7.(2) Pollen viability decreased gradually over time at different storage temperatures, declined rapidly at room temperature,and kept best at-20℃.The pollen viability of No.42 clone was up to 39.28% at-20℃for 14 days.(3) The best medium for five M. macclurei clones pollen was sucrose 100 g/L + boric acid 50 mg/L + agar 1%.
