摘要
目的:研究肿瘤相关中性粒细胞对人粘液表皮样癌MC3细胞上皮-间质转变(EMT)和侵袭转移过程调控的功能及机制。方法:分离培养C57小鼠骨髓中性粒细胞,Transwell检测肿瘤相关中性粒细胞对MC3侵袭和迁移的影响;Western印迹和Real-time PCR检测Vimentin等mRNA和蛋白表达;免疫荧光等检测Vimentin表达。结果:肿瘤相关中性粒细胞中TGF-β表达增加;MC3细胞与中性粒细胞共培养后Vimentin和Snail表达增加;裸鼠体内实验表明中性粒细胞浸润程度与Vimentin表达相关。结论:肿瘤相关中性粒细胞通过TGF-β诱导MC3细胞中Snail表达,并促进肿瘤侵袭和转移。
Objective:To investigate the function and mechanism of tumor-associated neutrophils in regulating EMT,invasion and metastasis of MC3 cells.Methods:Neutrophils were isolated from bone marrow of C57 mice and in vitro co-cultured with MC cells.Transwell and wound healing assays were performed to detect the effects of tumor-associated neutrophils on MC3 cell invasion and migration.mRNA and protein expressions of Vimentin and Snail in MC3 cells were detected by Western blot and real-time PCR assays.The distribution and effects of neutrophils on Vimentin expression were detected by immunofluorescence staining assay.Results:TGF-βexpression was increased in tumor-associated neutrophils.Tumor-associated neutrophils increased the expression of Vimentin and Snail in MC3 cells.Metastatic tumors were infiltrated by neutrophils and associated with Vimentin expression.Conclusion:Tumor-associated neutrophils can induce Snail expression by secreting TGF-β,and subsequently promote tumor cell EMT,invasion and metastasis of MC3 cells.
