摘要
目的:探讨何首乌提取物蒽醌、二苯乙烯苷对大鼠肝功能的影响。方法:选取SD大鼠80只,随机设为对照组、蒽醌组、二苯乙烯苷组和联合组,每组20只,在给药3个月、停药15d时每组大鼠随机处死10只,其中蒽醌组、二苯乙烯苷组和联合组分别给予1.1g/kg蒽醌、1.0g/kg二苯乙烯苷、1.1g/kg蒽醌联合1.0g/kg二苯乙烯苷进行灌胃,于给药3个月后、停药后15d检测肝功能、血清钙磷离子及肝脏指数。结果:联合组给药3个月后大鼠血清ALT、AST、ALP、钙离子、肝脏指数明显高于对照组、蒽醌组、二苯乙烯苷组(P<0.05),而LDH、磷离子明显低于对照组、蒽醌组、二苯乙烯苷组(P<0.05);对照组、蒽醌组、二苯乙烯苷组、联合组停药15d时ALT、AST、ALP、LDH、钙离子、磷离子及肝脏指数比较差异无统计学意义(P>0.05)。结论:何首乌中蒽醌、二苯乙烯苷对大鼠肝功能有损害作用,其中两者联合对肝功损害作用较大,但在停药后可恢复。
Objective: To investigate the effect of anthraquinone and stilbene glycoside from Polygonum multiflorum extract on liver function for rats.Methods: 80 SD rats were randomly divided into controlled group, anthraquinone group, stilbene glycoside group and combination group, 20 rats in each group, after 3 months of administration and 15 days of withdrawal, 10 rats in each group were killed at random, the anthraquinone group, the stilbene glycoside group and the combination group were given 1.1 g/kg anthraquinone, 1.0 g/kg stilbene glycoside, 1.1 g/kg anthraquinone combined with 1.0 g/kg stilbene glycoside, the liver function, serum calcium and phosphorus ions and liver index were measured at 3 months after administration and 15 days after withdrawal.Results: 3 months after administration, the serum ALT,AST,ALP,calcium ion and liver index in the combined group were significantly higher than those in the controlled group, the anthraquinone group and the stilbene glycoside group(P < 0.05),while LDH and phosphorus ions were significantly lower than those in the controlled group, the anthraquinone group and stilbene glycoside group(P < 0.05).There were no significant difference in ALT,AST,ALP,LDH,calcium, phosphorus and liver index in the controlled group, the anthraquinone group, the stilbene glycoside group and the combination group at 15 days after drug withdrawal(P > 0.05).Conclusion: Anthraquinone and stilbene glycoside in Polygonum multiflorum have impaired liver function in rats, the combination of them has greater impairment on liver function, but it can recover after drug withdrawal.
