基于大数据方法分析丹参酮IIA对乳腺癌免疫细胞浸润模式的影响 被引量:1

Analysis of the Effect of Tanshinone IIA on the Infiltration Pattern of Immune Cells in Breast Cancer Based on Big Data Method

摘要 目的运用大数据方法分析丹参酮IIA对乳腺癌浸润模式的影响。方法从相应数据库中分别下载乳腺癌组织和正常组织的转录组数据,以及丹参酮IIA作用于人乳腺癌细胞MCF-7的表达谱数据。对数据进行单样本基因集富集分析、生存分析、线性相关性分析、GO和KEGG富集分析。结果在乳腺癌组织中,有8种免疫细胞浸润与生存相关。丹参酮IIA作用于乳腺癌细胞的差异表达基因与6种免疫细胞浸润有较强相关性。丹参酮IIA调节乳腺癌免疫浸润的靶基因主要富集于细胞因子和细胞因子受体相互作用通路。结论丹参酮IIA可能通过相关基因和通路影响乳腺癌组织免疫细胞浸润模式,为进一步研究提供方向和依据。 Objective To analyze the effect of tanshinone IIA on the immune infiltration pattern of breast cancer using big data method.Methods The transcriptome data breast cancer tissues and normal breast tissues are downloaded from the University of California Santa Cruz(UCSC)Xena database,and the expression profile data of breast cancer cell line MCF-7 treated with tanshinone IIA from Gene Expression Omnibus(GEO)database.These data are then used to undertake the single sample gene set enrichment analysis(ssGSEA),K-M survival analysis,Correlation analysis,gene ontology(GO)analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis.Results There are eight kinds of immune cell infiltration related to survival in breast cancer tissues.Correlation analysis show that the differentially expressed genes of tanshinone IIA regulating breast cancer cells has a strong correlation with the infiltration of six types of immune cells.Through GO analysis and KEGG analysis,it is find that the target genes of tanshinone IIA regulating breast cancer immune infiltration is enriched in the pathway of cytokine-cytokine receptors interaction.Conclusion Tanshinone IIA may affect the immune cell infiltration pattern of breast cancer tissues through related genes and pathways,which provides direction and basis for further research.
