牦牛FSHB基因的克隆及生物信息学分析 被引量:1

摘要 通过PCR技术以及生物信息学软件,对牦牛FSHB基因进行克隆和生物信息学分析。结果表明:牦牛FSHB基因的c DNA序列区长645 bp,编码的氨基酸129个;同源性和分子进化树分析结果显示,其CDS区碱基序列与普通牛的关系最近,然后依次是水牛、山羊、绵羊、梅花鹿、马、人和小家鼠。牦牛FSHB蛋白的分子量14683,分子式C637H978N170O193S18,等电点6.27,总原子数1996,半衰期估计30 h,不稳定指数45.8,脂肪指数61.9;该蛋白属于非跨膜蛋白,α螺旋、延伸链、β转角和无规卷曲分别为30、29、6和64个,所占的比例分别为23.26%、22.48%、4.65%和49.61%。该基因编码的氨基酸在第18位点和第19位点可能存在信号肽。该研究探明牦牛FSHB基因与其他种属的差异,为进一步研究牦牛FSHB基因结构、功能及其与繁殖功能的关系提供了理论基础。 The FSHB gene of yak and its bioinformatics was cloned and analysed by PCR and bioinformatic software.The results showed that the c DNA sequence of yak FSHB gene was 645 bp,encoding 129 amino acids.The homology analysis and molecular evolutionary tree showed that the base sequence of CDS of yak FSHB gene was closely related to cattle,followed by buffalo,goats,sheep,the sika deer,horses,and mice.The molecular weight of yak FSHB was 14683,with C637 H978 N170 O193 S18 of the molecular formula,6.27 of the isoelectric point,1996 of the total atomic number,30 h of the half-life,45.8 of the instability index,and 61.94 of the fat index.The protein was a non-transmembrane protein.Theαhelix,extension chain,βturning angle,random coil of these protein are 30(23.26%),29(22.48%),6(4.65%)and 64(49.61%),respectively.It has a signal peptide at the 18 th and19 th sites.These results revealed the difference of FSHB gene between yak and other species,and gave the cues to explore the structure,function of FSHB gene and its relationship with reproductive function in yak.
