针康法对缺氧缺血性脑损伤新生大鼠神经功能及细胞凋亡相关蛋白表达的影响 被引量:4

摘要 目的?观察针康法不同时间介入对缺氧缺血性脑损伤新生大鼠神经功能及大脑皮质内Akt、14-3-3蛋白表达和细胞凋亡的影响。方法?将7日龄新生Wistar大鼠随机分成A组(假手术组)、B组(模型组)、C组(针刺组)、D组(康复组)和E组(针康组),每组再分为24 h、72 h和7 d 3个亚组。B组、C组、D组和E组用Rice-Vannucci法制备缺氧缺血性脑损伤模型。A组、B组不予任何治疗;C组进行头穴丛刺疗法,留针2 h;D组采取转笼、转棒方法,每次10 min;E组同时进行C组、D组2种治疗。结果?与A组相比,B组、C组、D组和E组Bederson评分、细胞凋亡数在各时间点均具有极显著差异(P<0.01),Akt、14-3-3蛋白的表达差异均具有统计学意义(P<0.05);C组、D组、E组在各时间点与B组比较,其Bederson评分均显著减少(P<0.05),其凋亡细胞数均明显减少(P<0.01),Akt、14-3-3蛋白的表达均明显增高(P<0.05);与E组比较,C组、D组Bederson评分在72 h和7 d时间点上均显著减少(P<0.05),其细胞凋亡数在各时间点差异均有统计学意义(P<0.05),Akt、14-3-3蛋白的表达在各时间点均显著提升(P<0.05)。结论?针康法可保护缺氧缺血性脑损伤新生大鼠的神经功能,减轻脑损伤,可能与促进脑细胞中Akt、14-3-3蛋白的表达,从而抑制细胞凋亡有关。 Objective To investigate the effect of different time intervention of acupuncture-rehabilitation therapy on the neurological function,the expression of Akt,14-3-3 proteins and apoptosis in the cerebral cortex of neonatal rats with hypoxic-ischemic brain damage.Methods 7-day-old newborn Wistar rats were randomly divided into groups A(sham operation),B(model),C(acupuncture),D(rehabilitation)and E(acupuncture-rehabilitation).Each group was divided into 3 subgroups at 24 h,72 h and 7 d.Rice-Vannucci method was used to establish the model of hypoxic-ischemic brain damage for B,C,D and E groups.The Groups A and B received no treatment;the group C treated with cluster needling of scalp acupuncture and the needle was kept for 2 hours;the group D adopted the method of rotating cage and rod for 10 minutes each;the group E received both acupuncture and rotating cage and rod training.Results Compared with group A,the Bederson scores and apoptosis number of B,C,D and E groups were significantly different at each time point(P<0.01),the expression of Akt and 14-3-3 protein was statistically significant at all time points(P<0.05).Compared with group B,the Bederson scores of group C,D and E was significantly lower(P<0.05),the number of apoptosis was significantly lower(P<0.01),the expression of Akt and 14-3-3 protein was significantly increased at all time points(P<0.05).Compared with group E,the Bederson scores of group C and D was significantly reduced at the 72 h and 7 d time point(P<0.05),the number of apoptosis was significantly different at each time point(P<0.05),the expression of Akt and 14-3-3 protein was significantly increased at all time points(P<0.05).Conclusion Acupuncture-rehabilitation therapy can protect the neurological function and alleviate the brain injury of the newborn rats with hypoxic-ischemic brain damage,which may be related to promoting the expression of Akt and 14-3-3 protein in brain cells,thereby inhibiting the apoptosis.
