摘要
目的探讨天麻醇提取物对帕金森病(PD)大鼠认知功能障碍的改善作用及其机制。方法采用两点法分别于中脑腹侧被盖部和黑质致密部注射6-羟基多巴胺(6-OHDA)复制大鼠PD模型。取PD模型复制成功大鼠随机分为天麻醇提取物低剂量组、天麻醇提取物中剂量组、天麻醇提取物高剂量组、美多巴组、模型对照组,另取正常大鼠作为空白对照组。天麻醇提取物低、中、高剂量组分别灌胃给予5 g生药/kg、10 g生药/kg、20 g生药/kg的天麻醇提取物,美多巴组灌胃给予7.8 mg/kg的美多巴,空白对照组和模型对照组灌胃相同体积的蒸馏水,1次/d,连续21 d。以盐酸去水吗啡0.5 mg/kg腹腔注射诱导大鼠旋转,观察注射5 min后30 min内大鼠恒定向健侧旋转的转圈数;以Morris水迷宫实验[大鼠在平台所在象限的距离百分比(PT%)和停留时间百分比(T%)]评价天麻醇提取物对PD大鼠认知功能障碍的改善作用;以光学法测定血清脂质过氧化物(LPO)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA),评价天麻醇提取物的抗氧化能力。结果与模型对照组比较,天麻醇提取物低剂量组、中剂量组和高剂量组PD大鼠转圈数均有降低趋势,但P均>0.05;天麻醇提取物高剂量组在水迷宫的PT%和T%增加(P均<0.05)。与空白对照组比较,模型对照组血清中LPO、MDA含量升高,血清GSH-Px、SOD含量降低(P均<0.01);与模型对照组比较,天麻醇提取物高剂量组大鼠血清中LPO含量降低(P<0.05),天麻醇提取物低剂量组、中剂量组和高剂量组血清中GSH-Px、SOD含量升高、MDA含量降低(P<0.05或<0.01)。结论天麻醇提取物能明显改善PD大鼠的认知功能障碍,其机制可能是增强了机体抗氧化能力。
Objective To study the improvement effect of gastrodia alcohol extract on cognitive dysfunction in Parkinson’s disease(PD)rats and and antioxidant mechanism.Methods A two-point method was used to inject 6-hydroxydopamine(6-OHDA)into ventral tegmental area(VTA)and substantia nigra pars compacta(SNc)of rats to copy the models of PD.The rats that successfully replicated the PD models were randomly divided into the model group,the medopa group,the low-dose,medium-dose and high-dose groups of Gastrodia alcohol extract,and meanwhile,some normal rats were taken as the normal group.Among them,the low-dose,medium-dose and high-dose groups of Gastrodia alcohol extract were given 5,10,and 20 g crude drug/kg by gavage,respectively,and the rats in the medopa group were given 7.8 mg/kg of medopa by gavage.The rats in the control and model groups were given the same volume of distilled water.Gastric perfusion was done once per day,continuously for 21 days.The rat rotation was induced by intraperitoneal injection of0.5 mg/kg apomorphine hydrochloride(APO),and the number of constant rotation to the healthy side was recorded within30 min after 5 min of injection.Morris water maze experiment[the percentage of distance(PT%)and time(T%)that rats stay in the quadrant where the platform is located]was used to evaluate the improvement effect of Gastrodia alcohol extract on the cognitive dysfunction in PD rats.The serum levels of SOD,MDA,GSH-Px,and LPO were measured by optical method to evaluate the antioxidant effect of gastrodia alcohol extract.Results Compared with the model group,the number of rotation of PD rats in the low-dose,medium-dose and high-dose groups of Gastrodia alcohol extract had a decreasing trend,but no significant difference was found(P>0.05).In the high-dose group of Gastrodia alcohol extract,PT%and T%in morris water maze experiment increased significantly(both P<0.05).Compared with the control group,the serum levels of LPO and MDA in the model group increased,and the SOD and GSH-Px decreased(all P<0.01).Compared with the model group,the serum content of LPO in the high-dose group of Gastrodia alcohol extract was reduced(P<0.05).The activity of GSH-Px in the low-dose,medium-dose and high-dose groups of Gastrodiae alcohol extract increased,the serum content of SOD increased,and the serum content of MDA decreased(P<0.05 or P<0.01).Conclusion Gastrodia alcohol extract can significantly improve the cognitive dysfunction of PD rats,and its mechanism may be related to the enhancement of antioxidant capacity of the body.
