miR-432通过下调HOXC13表达抑制胶质瘤细胞的增殖、迁移和侵袭 被引量:1

miR-432 inhibits cell proliferation,migration and invasion of glioma by targeting HOXC13

摘要 目的研究miR-432对胶质瘤细胞株U251细胞增殖、迁移和侵袭的影响及潜在的分子机制。方法采用miR-432寡聚核苷酸(miR-432 mimics)转染U251细胞;用实时定量PCR(RT-qPCR)检测转染后的细胞中miR-432的表达水平;用CCK-8法测定细胞增殖活性,Transwell实验检测细胞迁移和侵袭能力,双荧光素酶报告系统验证miR-432与HOXC13的调控关系。结果与正常人脑胶质组织及胶质细胞株相比,胶质瘤组织和胶质瘤细胞系中的miR-432表达水平均显著降低(P<0.05~0.01);过表达miR-432可抑制U251细胞增殖、迁移和侵袭,并抑制HOXC13的蛋白和mRNA表达。双荧光素酶报告系统结果也表明,miR-432靶向调控HOXC13的表达;HOXC13在胶质瘤中高表达,并促进胶质瘤细胞的增殖、迁移和侵袭;过表达miR-432可拮抗胶质瘤中HOXC13促进细胞增殖、迁移和侵袭的作用。结论miR-432通过靶向负调控HOXC13基因抑制胶质瘤细胞的增殖、迁移和侵袭,miR-432是胶质瘤的潜在治疗靶点。 Objective To investigate the effect of miR-432 on the proliferation,migration and invasion of glioma cell line U251 and its underlying molecular mechanism.Methods U251 cells were transfected with miR-432 mimics,RT-qPCR was used to detect miR-432 expression in transfected cells,CCK-8 was used to determine cell proliferation activity,Transwell assay was used to detect cell migration and invasion ability and the dual luciferase reporter system was used to verify the regulatory relationship between miR-432 and HOXC13.Results Compared with normal human brain tissues,miR-432 expression significantly reduced in glioma tissues(P<0.05).Overexpression of miR-432 could inhibit U251 cell proliferation,migration and invasion and inhibit HOXC13 protein and mRNA expression.Dual luciferase report system results showed that miR-432 targets with HOXC13.HOXC13 was highly expressed in gliomas and promoted glioma cell proliferation,migration and invasion.Overexpression of miR-432 can antagonize the effect of HOXC13 on cell proliferation,migration and invasion in gliomas.Overexpression of Mir-432 can antagonize the effect of HOXC13 in glioma on promoting cell proliferation,migration and invasion.Conclusions miR-432 inhibits the proliferation,migration,and invasion of glioma cells by targeting the HOXC13 gene.miR-432 is a potential therapeutic target for gliomas.
